[0022] Example 1: Effect of allicin on the stability of lipid rafts in mouse mammary gland tissue

[0023] 1. Materials and methods

[0024] 1.1 Experimental materials:

[0025] The mice used were adult postpartum female BALB / c mice, a total of 60 (8-10 weeks old, body weight 25-30 g). All mice were provided by Hubei Experimental Animal Center. All animal experiments were performed in accordance with the Guide for the Care and Use of Laboratory Animals of the National Institutes of Health and approved by the Institutional Animal Care and Use Committee of Huazhong Agricultural University.

[0026] Allicin (purity>99.8%) was purchased from National Institute of Pharmaceutical and Biological Products Control (Beijing), and dissolved in normal saline.

[0027] Staphylococcus aureus (ATCC 35556) was purchased from American Type Culture Collection (Manassas, VA, USA).

Abstract

The invention discloses application of allicin in reduction of stability of lipid rafts. It is discovered that a certain dose of allicin can inhibit formation of the lipid rafts in the mammary gland tissue of a mouse and / or lower the content of the lipid rafts for the first time, and accordingly the stability of the lipid rafts in mammary gland cells is effectively lowered. Therefore, the allicincan be used as a novel inhibitor for the lipid rafts, and the technical support is provided for development of a medicine for treating the diseases related to the lipid rafts.

Description

technical field [0001] The invention relates to the technical field of medical application of allicin, in particular to the application of allicin in reducing the stability of lipid rafts. Background technique [0002] Lipid raft is a liquid ordered phase dynamic micro-domain rich in cholesterol and sphingomyelin, with a diameter of 50-350nm. phase, such as the phase separation formed by the liquid crystal phase and the gel phase. Lipid rafts are microdomains containing specific esters and proteins. Lipid rafts are mainly composed of cholesterol, sphingolipids and some special proteins. Sphingolipids are a class of complex compounds with sphingosine as the backbone, mainly divided into sphingomyelin, glycosphingolipid and ceramide. Among them, sphingomyelin is mainly composed of fatty acids, sphingosine and phosphorylcholine; glycosphingolipids are mainly composed of fatty acids, sphingosine and sugar; ceramide, as a special form of sphingomyelin, also plays an important ...
