The invention discloses a glutathione reductase detection reagent kit which is prepared from the following components: a first reagent and a second reagent. The first reagent is prepared from the components of a phosphate buffer solution, EDTA-NA2, ascorbic acid oxidase, potassium ferricyanide, a preservative and oxidized glutathione. The second reagent is prepared from the components of a trimethylamine methane buffer solution, EGTA, a preservative and reduced coenzyme. The first reagent and the second reagent are both liquid and can be stored for 12 months at 2-8 DEG C environment. Potassiumferricyanide can oxidize hemoglobin to produce stable hemiglobincyanide, and the influence of detection results is reduced. The second reagent adopts EGTA, the linear relationship between the detection results and the change rate of absorbance is can be improved, and adopting of two-point calibration is convenient. The invention further discloses application using the glutathione reductase detection reagent kit.
