The invention discloses a method for marker-assisted selection of growth traits by using cattle MOGAT1 gene SNP markers and an application of the method. The method comprises the following steps: withcattle whole genome DNA as a template, amplifying partial fragments of the MOGAT1 gene by using specific primers, digesting a PCR product by using restriction enzyme TaqI, and carrying out agarose gel electrophoresis on the fragment subjected to enzyme digestion; and identifying the genotype of the cattle MOGAT1 gene mutation site g.25940T > C according to an agarose gel electrophoresis result. According to the invention, a PCR-RFLP method is used for DNA level typing detection; SNP markers closely related to the growth traits like cattle weight are found; the SNP markers can be used for marker-assisted selection of the growth traits; and cattle specialized meat populations with excellent genetic resources are rapidly established.
Description
technical field [0001] The invention belongs to the field of molecular genetic breeding and relates to a method for detecting the single nucleotide polymorphism (SNP) of the mutation site g.25940T>C of the cattle MOGAT1 gene. Background technique [0002] Currently, a variety of molecular markers have been widely used, including single nucleotide polymorphism (SNP) and so on. SNP technology has the following advantages: (1) The number of SNPs is huge and distributed on the whole genome; (2) The inheritance of SNPs is stable, and the probability of mutation is low, which also makes SNPs have a role in molecular marker-assisted selection. (3) Most of the SNPs belong to two alleles, which is beneficial to determine the genotype of individuals in a large-scale population. [0003] PCR-RFLP is a commonly used method in SNPs detection. The principle of this method is to use the specificity of the restriction endonuclease enzyme cutting site to act on the same DNA fragment, an...
