Embodiment 1

[0033] I. MATERIALS AND METHODS

[0034] 1. TC6A database: The ZSCAN16 expression profiles of 19 paired cancers and adjacent normal TCGA samples and the clinical information of bladder cancer samples were obtained from the TCGA database. The clinical data of the 19 patients are shown in Table 1.

[0038] 2. Cell Culture and Testing

[0039] 2.1 Cell Culture and Reagents

[0040] First, two human bladder cancer cell lines (T24 and 5637) were selected, both purchased from ATCC, and were cultured and passaged according to the instructions. Briefly described as follows: T24 and 5637 cells in 5x10 4 / ml concentration was inoculated in a T-25 petri dish containing 10ml RPMI-1640 (purchased from Gibco Co., Ltd.) + 10% (v / v) FBS (purchased from Sigma Aldrich Co., Ltd.) culture medium, and placed at 37°C, Cultured in a 5% carbon dioxide incubator. After verifying the silencing efficiency of ZSCAN16 by qPCR (re...

[0081] Example 2: Animal experiment

[0082] 1. Materials and methods

[0083] 4-week-old female nude mice were purchased from Shanghai Lingchang Biotechnology Co., Ltd., the body weight and size were roughly the same before the experiment, and they were randomly divided into 2 groups, 10 mice in each group, and 1*107 non-silencing siRNA T24 cell lines were taken from each (implementation The T24ZSCAN16 NC group of Example 1), and the lentivirus-transfected and anti-ZSCAN16 shRNA-transduced T24 cell lines (T24 ZSCAN16 KD2 group of Example 1) were injected subcutaneously into the right armpit of nude mice, respectively, and the average body weight of each group The difference was not statistically significant (P2 ) measurement, and then sacrificed for measurement.

[0084] 2. Results

[0085] See Figures 5A-5G Shown: In the in vivo experiment of the nude mouse bladder cancer model, the experimental group and the control group were subcutaneously injected with shZSCAN16 (KD2...

Abstract

The invention relates to a novel tumor-related transcription factor ZSCAN16 and its function in inhibiting tumors, belonging to the technical field of biomedicine. Application of ZSCAN16 gene as a therapeutic target for bladder cancer. The advantages of the present invention are as follows: the present invention reveals that ZSCAN16 may play an important biological role in the development of bladder cancer, and it is also closely related to the development of tumors. The results of the present invention show that ZSCAN16 is a key new oncogene for the occurrence and development of bladder cancer. The results of in vitro experiments showed that silencing of ZSCAN16 inhibited the proliferation, colony formation, apoptosis, migration and invasion of T24 cells. ZSCAN16 can be used as a research target for bladder cancer regulation, as a tumor diagnosis and prognosis evaluation marker, and as a target to develop tumor-inhibiting drugs.

Description

technical field [0001] The invention relates to a novel tumor-related target ZSCAN16 and its application in inhibiting tumors, and belongs to the technical field of biomedicine. Background technique [0002] Bladder cancer is one of the most common urogenital malignancies in the world. Since 2015, a total of 3.4 million people worldwide have suffered from bladder cancer, with 430,000 new cases every year. The incidence and mortality of bladder cancer in China have risen rapidly over the past decade. Smoking, family history, radiation therapy, recurrent bladder infections, and chemical exposure have been shown to be high-risk factors for the development and progression of bladder cancer. Standard treatments for patients with bladder cancer include radiation therapy, chemotherapy, and surgical resection. The treatment of non-muscle invasive bladder cancer includes endoscopic bladder tumor resection, postoperative adjuvant intravesical chemotherapy, and commonly used drugs in...
