Problems solved by technology

However, this test is primarily performed when an underlying birth condition (such as PPROM) occurs and may not be available in women with intact membranes
In other words, current diagnostic tests may not be able to predict potential preterm birth, such as PPROM

Embodiment 1

[0147] Example 1 - Prediction of preterm birth status

[0148] In an example, a patient who is 6 months pregnant presents the following risk factors: low socioeconomic status, past history of bleeding during her pregnancy, and history of preterm delivery during previous pregnancies. Physicians need to identify the likelihood of preterm birth in a patient and suggest using the methods and systems provided herein to predict the likelihood of preterm birth. Obtain a sample of the patient's vaginal fluid for analysis of the vaginal microbiome. The vaginal sample is processed to generate data indicative of the distribution of different types of microbial populations in the vaginal sample. Algorithms are trained to identify different types of microorganisms, such as Lactobacillus, Escherichia, Prevotella, Enterococcus, Candida genus, staphylococcus and herpes virus. The trained algorithm predicted that the subjects had an approximately 88 percent risk of preterm birth. The train...

Embodiment 2

[0149] Example 2 - Prediction of Preterm Birth Risk

[0150] In this example, four pregnant women (ie, subjects #1-4) who showed signs of threatening preterm birth at different time points in pregnancy were assessed for preterm birth risk by the method of the present invention. Specifically, vaginal fluid samples were obtained from each subject and processed as described in Example 1. As shown in Example 1, the risk of a subject's preterm birth condition was predicted using the trained algorithm with an accuracy of 98.15%. Table 3 shows the predicted probability of preterm birth condition (PBC) and predicted birth outcomes based on the analysis of the microbial population in the vaginal samples, as well as data on the actual birth outcomes for each subject.

Abstract

Methods and systems (301) are provided to predicting premature birth condition in a subject. The method for predicting or monitoring premature birth condition in a subject comprises processing a biological sample obtained from the subject to generate data indicative of a distribution of a plurality of populations of microbes of different types in the biological sample. A presence, absence, or relative amount of an individual population of the plurality of populations of microbes may be indicative of a premature birth condition. Next, a trained algorithm may be used to process the data to determine a presence, absence, or relative amount of the individual population of microbe. Next, based on the presence, absence, or relative amount, the subject may be identified as having the premature birth condition, such as, for example, in a report.

Description

[0001] priority statement [0002] This application claims priority from PCT application PCT / CN2018 / 112965 filed on October 31, 2018, the entire contents of which are incorporated herein by reference. Background technique [0003] Preterm birth is the leading cause of death among children under 5 years of age worldwide and is the leading cause of perinatal morbidity and mortality. In 2015, preterm birth and low birth weight accounted for about 17 percent of infant deaths. In the United States, 10 percent of babies are born prematurely each year. One-third of all preterm or preterm births are caused by preterm premature rupture of membranes (PPROM). Spontaneous rupture of membranes (ROM), ie rupture of the amniotic sac, is a normal part of labor and delivery of the fetus. Premature rupture of membranes (PROM) is the rupture of the membranes before labor begins, regardless of gestational age. When PROM occurs at term, labor is usually spontaneous or induced within 12-24 hour...
