Embodiment 1

[0047] Embodiment 1 research object

[0048] In this study, from January 2019 to June 2021, a total of 200 cases of medical records were collected by the prenatal diagnosis laboratory, divided into preeclampsia (PE) group and normal control group, and the case data, placental tissue samples, venous blood.

Embodiment 2

[0049] The collection of embodiment 2 placental tissue samples

[0050] Immediately after the placenta was delivered by cesarean section, two pieces of placenta with a size of about 1cm×1cm×1cm and a little decidua tissue were taken from the center of the mother’s surface of the placenta under aseptic condition, and the specimens were placed in a sterile Eppendorf tube and placed in liquid nitrogen for 15 minutes. Save until RNA extraction. Real-time PCR was used to detect the miR-27a-5p level in the placental tissue of each group.

Embodiment 3

[0051] The collection of embodiment 3 venous blood

[0052] Pregnant women draw 5ml of venous blood before giving birth and collect it in EDTA vacuum tube. The extracted venous blood was centrifuged in a centrifuge at 4° C. and 1500 g for 15 minutes within 30 minutes, and then the plasma was separated for RNA extraction.

Abstract

The invention provides a method for diagnosing whether a subject has preeclampsia or is at the risk of preeclampsia, and further provides application of microRNA and / or a specific probe / detection primer of the microRNA in preparation of a diagnostic composition and the diagnostic composition for detecting preeclampsia. The expression of the microRNA 27a in the plasma in a preeclampsia patient is obviously reduced, and the microRNA 27a can be used as a diagnostic index which is valuable, simple, convenient and easy to dynamically monitor preeclampsia; and meanwhile, the reduction of the microRNA 27a in the placenta tissue is further a potential diagnostic index for predicting preeclampsia.

Description

technical field [0001] The invention belongs to the technical fields of biomedicine and molecular biology, and in particular relates to the application of peripheral blood microRNA 27a in the diagnosis and treatment of preeclampsia. Background technique [0002] Pre-eclampsia (Pre-eclampsia, PE) is a hypertensive disorder that occurs during pregnancy, affecting about 5-8% of pregnant women, and PE / eclampsia is related to 10-15% of maternal deaths directly. PE is associated with eclampsia and HELLP syndrome in the mother, as well as preterm birth, intrauterine growth restriction, and perinatal death in the fetus. PE has adverse effects on maternal and offspring health and quality of life after delivery. Women with a history of PE have a 2-fold increased risk of cardiovascular complications later in life and a 5- to 12-fold increased risk of end-stage renal disease. Preeclampsia can be subdivided into early-onset (EOPE) and late-onset (LOPE) phenotypes based on diagnosis bef...
