Method for extracting carrageenan

The invention discloses a method for extracting carrageenan, which comprises the following steps: washing a raw material with water, performing combined alkali treatment on the raw material, wherein the concentration of alkali is decreased gradually, washing the raw material to enable the pH to be 7-8; carrying out composite enzyme treatment, removing heavy metals by using a metal chelating agent, boiling carrageenan, filtering coarsely, filtering finely, reducing temperature, gelling, dewatering, cutting into strips, drying, crushing and screening so as to obtain the carrageenan. According to the invention, a mixed solution of NaOH, KOH and LCI with gradually decreased concentration in three grades is adopted in an alkali treatment process, the loss of carrageenan is reduced effectively while the alkali treatment time is reduced; cellulase and protease combined treatment is adopted, and the yield of the carrageenan is improved while the high gel strength of the carrageenan is ensured; ethylene diamine tetraacetic acid and nopotassium phosphate are adopted for further treatment for the raw material, and the heavy metals in the raw materials are removed effectively. The gel strength of the carrageenan prepared by the method is greater than or equal to 2139.2 g / cm<2>, the extraction rate is greater than or equal to 23.9 percent, Pb is less than or equal to 2.5 mg / kg, As is less than or equal to 1.0 mg / kg, Hg is less than or equal to 0.5 mg / kg, and the Cd is less than or equal to 1.0 mg / kg.

Ethanol yield and reduction of biomass accumulation in the recombinant strain of Saccharomyces cerevisiae overexpressing atpase

A new approach for increase of ethanol yield during alcoholic fermentation via decrease of biomass accumulation by using ATP degrading enzymes is described. The part of the Saccharomyces cerevisiae SSB1 gene coding for cytosolic ATPase domain of ribosome associated chaperon cloned into expression cassette under control of the glycerol-3-phosphate dehydrogenase gene (GPD1) promoter was introduced into the S. cerevisiae BY4742 strain. The recombinant strains were tested for their ability to grow and produce ethanol during glucose anaerobic and aerobic cultivations. Strains overexpressing ATPase domain of SSB1 possessed decreased concentration of intracellular ATP. They accumulated elevated amounts of ethanol and were characterized by decreased biomass accumulation as compared to the wild-type strain under both anaerobic and aerobic conditions. Similarly, the apyrase gene apy from E. coli encoding ATP / ADP hydrolyzing phosphatase and ATPase domain of SSB1 gene of S. cerevisiae were co-expressed under the control of galactose-inducible GAL1 promoter. The recombinant S. cerevisiae strains revealed slight reduction of biomass accumulation, while specific ATPase activity, ethanol accumulation and yield during alcoholic galactose fermentation under semi anaerobic conditions were increased.

The present invention discloses a cyclic utilization method of solar silicon wafer texturing solution. The method comprises the following steps of: (1) performing etching processing of a solar siliconwafer by employing a texturing solution, employing a reverse osmosis membrance circulation separation device to separate fluosilicic acid impurities from the mixed solution after etching on an etching groove; (2) performing quantitative analysis of hydrofluoric acid and nitric acid in the mixed solution after separation of the impurities respectively by employing an automatic measurement potentiometric titrator and a nitrate ion selective electrode automatic measurement potentiostat to obtain component concentration increment or decrement of the mixed solution after etching; and (3) accordingto the component concentration increment or decrement, adding acid with decreased concentration into the etching process or the cyclic utilization to retake as the etching solution of the silicon wafer. The effectively utilization of the mixed waste acid solution discharged in the etching process allows the economical benefit to be remarkable, greatly reduces the discharge of the mixed waste acidsolution with strong acid components and have remarkable environmental benefit.
