Embodiment Construction

[0024] A rotary platform for cell lysis, sample purification and elution is disclosed herein. Use of the platform enables simplified movement of magnetic particles or beads and associated sample fragments such as nucleic acids through a multi-stage cell lysis and purification protocol without the need for external pipetting systems and associated pipetting tips.

[0025] A rotating tray with at least one sequence of treatment recesses distributed along its perimeter is mounted for selective rotation and elevation relative to the extendable and retractable magnets. A magnet enables the magnetic beads to be selectively attracted into clusters within the recess. The rotating and raising movement of the tray enables the magnetic bead clusters to move from well to well without the use of a pipetting system or associated disposable tips, while the magnet remains stationary. The system may also include extendable and retractable agitators to selectively agitate the contents of the w...

Abstract

Magnetic beads having cell components of interest are translated between a sequence of processing wells in a tray without need for pipetting. The circular tray contains one or more sequences of wells each interconnected by a respective channel. The tray is rotated about a central axis and a magnet, an agitator, and a heater provided external to the tray enable magnetic bead translation, mixing, and incubation, respectively. The magnet proximate a well forms a cluster of beads. Manipulation of the tray in rotation and elevation results in translation of the cluster from one well, through a channel, and into an adjacent well. The well containing a cluster may be rotationally positioned in front of the agitator, the agitator extended into contact with the well, followed by mechanical agitation. The heater, disposed beneath the tray, may accept a well lowered thereto for selective heating.

Description

technical field [0001] The disclosure herein is generally in the field of cell lysis, purification and elution. More particularly, the present disclosure relates to a rotating multi-recess tray with particular utility in the field of cell lysis, purification and elution in molecular diagnostics. Background technique [0002] In a typical cell lysis and purification system, magnetic beads used to capture desired sample components such as DNA and RNA are held in a well or some other processing volume while unwanted sample fluid is removed from the well, And wash buffer and other buffers are added to the wells by pipetting system. External magnets capture and hold the magnetic beads during the fluid removal process. When another fluid is reintroduced into the well, the beads are usually released from the influence of the magnet to achieve turbulent mixing. [0003] An alternative approach practiced in the prior art is to move the magnetic beads and associated target componen...
