Embodiment 1

[0034] Early morning fasting serum samples were collected from pregnant women with preeclampsia and healthy pregnant women during pregnancy or on the day of delivery, aliquoted and stored at -80°C until the day of testing.

[0035] Human BMP6 ELISA kit (LSBio, #LS-F4538) was used to perform ELISA assay on the collected serum samples from pregnant women with preeclampsia and healthy pregnant women. Specifically, the detection was performed according to the manufacturer's instructions of the ELISA kit. On the SpectraMax Plus microplate reader of Molecular Devices, USA, subtract the reading at 540nm from the reading at 450nm to obtain the final absorbance value.

[0036] The serum BMP6 level of the above samples can be calculated by adding the absorbance value into the standard curve fitting formula (implemented specifically according to the instructions of the ELISA kit manufacturer).

[0037] The results showed that serum BMP6 levels were significantly higher in patients with ...

Embodiment 2

[0041] Human trophoblast cells were treated with human BMP6 protein (50ng / ml) (R&D, #355-BM-010) or blank vector for 24 hours.

[0042]The cells treated in step 1 were respectively digested and resuspended in DMEM medium containing 0.1% (vol / vol) fetal bovine serum, and 250 μl containing 8x10 4 The cell suspension of two cells was inoculated in the treated Transwell chamber (the Transwell chamber had been coated with 40 μl Matrigel at a concentration of 1 mg / ml in advance, the Transwell chamber had a pore size of 8 μm, and both the Transwell chamber and Matrigel were purchased from BD Biosciences ). Add 750 μl of DMEM medium containing 10% (vol / vol) fetal bovine serum to the lower layer of the Transwell chamber. After the inoculated cells were incubated at 37°C for 36h, the non-invaded cells on the upper part of the membrane were wiped clean, and the cells on the lower part of the membrane were fixed with cold methanol (-20°C) and air-dried. The nuclei were stained with Hoec...

Abstract

The invention relates to the technical field of biological medicine, in particular to application of bone morphogenetic protein 6 in early diagnosis and treatment of preeclampsia. The invention provides application of BMP6 as a marker for early diagnosis/treatment of preeclampsia, application of BMP6 combined with PlGF and activin A in early diagnosis of preeclampsia, and application of BMP6 in preparation of drugs for prevention and treatment of preeclampsia. The invention finds that the BMP6, PlGF and activin A can improve the diagnosis effect of the existing diagnostic marker on preeclampsia in the preeclampsia early diagnosis; in addition, it is found that BMP6 can increase invasiveness and endothelial characteristic acquisition of human trophoblast cells, improve insufficient placental trophoblast invasion and poor vascular remodeling in the early stage of preeclampsia and then improve development of preeclampsia, and the BMP6 has the potential of preparing drugs for preventing and treating preeclampsia.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to the application of bone morphogenetic protein 6 in early diagnosis, prevention and treatment of preeclampsia. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present invention, and is not necessarily taken as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] Preeclampsia is a common pregnancy-related disorder and one of the leading causes of maternal and neonatal morbidity and mortality. Due to the complex etiology and pathogenesis of preeclampsia, there is still a lack of effective target therapy drugs in addition to symptomatic treatment in clinical diagnosis and treatment, and most patients have to accept surgical treatment for early termination of pregnancy. In addit...
