Example Embodiment

[0077] Example 1

[0078] A reagent for measuring reticulocytes with the following composition was prepared.

[0079] Fluorescent dye (III) 7mg

[0080] NaH 2 PO 4 .H 2 O 53.8mg

[0081] Na 2 HPO 4 .7H 2 O 163.4mg

[0082] Coco Betaine 100mg

[0083] Delicate water 1L

[0084] (adjust pH to 7)

[0085] 4 microliters of anticoagulant-treated blood was added to 1 milliliter of the reagent, and incubated in an incubation tank at 40° C. for 30 seconds to form a measurement sample.

[0086] The sample for measurement is detected by the method of the present invention and the detection device of the red semiconductor laser of the present invention. The excitation wavelength of the red semiconductor laser is 633nm-635nm, and the power is 5mW. By measuring the forward low-angle scattered light FSC intensity and the fluorescence FL intensity, the following figure 2 The scatter plot shown. The measurement results showed that the proportion of reticulocytes (RET) in the to...

[0100] Example 3:

[0101] To demonstrate the location of reticulocytes on the scatter plot, we purified reticulocytes from blood. A reagent for measuring reticulocytes with the following composition was prepared.

[0102] Fluorescent dye (III) 7mg

[0103] NaH 2 PO 4 .H 2 O 53.8mg

[0104] Na 2 HPO 4 .7H 2 O 163.4mg

[0105] Coco Betaine 100mg

[0106] Delicate water 1L

[0107] (adjust pH to 7)

[0108] Adopt the method identical with embodiment 1 to detect, the result is as follows Figure 4 as shown, Figure 4 Scatterplot for the detection of reticulocytes purified and isolated from whole blood. From the figure, we can clearly see the location distribution of higher concentration reticulocytes on the scatter plot.

Abstract

The invention provides a quick and cheap method of measuring the reticulocyte in a sample and a device capable of detecting the reticulocyte through the method. The method provided by the invention is capable of detecting the reticulocyte on a full automatic blood analyzer making use of the flowing type cell technology. The method adopts red fluorescent dye and takes red laser as an activation light source to complete the automatic detection of the reticulocyte within 60 seconds.

Description

technical field [0001] The invention relates to a cell detection method and device, in particular to a reticulocyte detection method and detection device. Background technique [0002] Reticulocytes are immature red blood cells between late immature red blood cells and mature red blood cells. The reticulocyte count is an important index to evaluate bone marrow hematopoietic function and erythropoietic activity. [0003] Under normal physiological conditions, it takes 4 days for the bone marrow to transform from primitive cells to reticulocytes, and then the reticulocytes mature in the bone marrow for 48 hours and enter the peripheral blood, and the reticulocytes in the blood continue to mature for 48 hours to become mature red blood cells. The number of reticulocytes in peripheral blood is of great significance for evaluating bone marrow erythroid hematopoiesis and the transfer rate of reticulocytes from bone marrow to peripheral blood [0004] Counting reticulocytes is a ...
