Problems solved by technology

There are many methods to improve the fermentative acid production capacity of succinic acid, but so far, under pure anaerobic conditions, there are few reports on strains that can efficiently use glucose as a single carbon source and produce high succinic acid

Embodiment 1

[0032] This example illustrates the Escherichia coli ( Escherichia coli ) BER108 (CCTCC M 2012068) for the first step of continuous culture acclimation.

[0033] Escherichia coli original bacterial strain carries out the method for first step continuous culture acclimatization as follows:

Embodiment 2

[0037] This embodiment illustrates that screening obtains good Escherichia coli (Escherichia coli ) BER208 method.

[0038] Screening steps:

[0039] 1. Solid plate primary screening

[0040] Under aseptic conditions, take out 2-4mL bacterial liquid from the continuous culture device and dilute it with sterile water 1×10 4 times, take 100 μL and spread it on a solid plate, incubate anaerobically at 37°C for 24 hours, and select colonies with fast growth and fullness.

[0041] 2. Solid flat plate re-screening

[0042] The screened strains were repeatedly transferred to the plate, and finally obtained the strains BER10806, BER208, BER10811 and BER10814, which showed strong growth rate and growth stability.

[0043] 3. Shake flask fermentation screening

[0044] The strains BER108, BER10806, BER208, BER10811 and BER10814 were inoculated into the seed medium for expanded cultivation, filled with carbon dioxide for 2 minutes, 37° C., 200 r / min, and cultivated for 12 hours. The...

Embodiment 3

[0053] This embodiment illustrates that screening obtains good Escherichia coli ( Escherichia coli ) Passaging stability of BER208.

[0054] On solid plates, Escherichia coli ( Escherichia coli ) BER208 was transferred to the point for multiple times, and the obtained strains were subjected to fermentation verification respectively. The experimental results are shown in Table 2:

[0055] Table 2 Escherichia coli ( Escherichia coli ) BER208 Passage Stability Analysis

[0056]

[0057] It can be seen from the experimental results that after 8 consecutive subcultures, Escherichia coli ( Escherichia coli ) BER208 has relatively stable succinic acid production and succinic acid conversion rate, and has good passage stability, and can be used as a production strain for further research and development.

Abstract

The invention discloses a strain capable of producing succinic acid, a method for producing the succinic acid and an application thereof. The strain is named as Escherichichia coil BER208 with the collection number of CCTCCNO: M2012351. The strain grows quickly with glucose serving as the single carbon source in a fermentation medium under the anaerobic condition and is capable of highly producing the succinic acid. The optical density (OD) 6000 reaches 6.87 and the yield of the succinic acid reaches 10.5g / L after fermentation for 48 hours, and compared with an original strain, the yield of the succinic acid is improved by more than two times.

Description

technical field [0001] The invention relates to the field of microorganisms, in particular to a succinic acid-producing bacterial strain and a method and application thereof for producing succinic acid. Background technique [0002] Succinic acid (also known as succinic acid) is a common natural organic acid widely found in the human body, animals, plants and microorganisms. As an important chemical raw material, succinic acid is widely used in industries such as medicine, food and surfactant. In recent years, with the depletion of fossil resources and the increasingly serious environmental problems, the use of biosynthesis instead of traditional chemical synthesis to prepare succinic acid has attracted much attention. The US Department of Energy listed succinic acid as the 12 most valuable substances in the future One of the biorefinery products. [0003] Biosynthesis of succinic acid is to use various microorganisms such as bacteria and fungi to produce succinic acid thr...
