specific Embodiment approach 1

[0020] Specific embodiment one: the method for quickly and efficiently separating wetland benthic invertebrates in this embodiment is realized in the following steps:

[0021] 1. Collect benthic invertebrate carrier samples in the wild, add ethanol solution with a volume concentration of 75% to 95% for preservation;

[0022] 2. Take out the benthic invertebrate carrier sample preserved in step 1, then add the solution, stir for 5 to 15 minutes and remove large particles of impurities during the stirring to obtain a mixed solution;

[0023] 3. The mixed solution obtained in step 2 is left to settle for 10 to 20 minutes to obtain the supernatant solution of the upper layer of the mixture and the sediment of the lower layer;

[0024] 4. Replace the benthic invertebrate carrier sample in step 2 with the sediment obtained in step 3, repeat the operation 3 to 5 times according to the method of steps 2 and 3, and obtain 4 to 6 parts together with the upper layer solution obtained in ...

specific Embodiment approach 2

[0036] Embodiment 2: This embodiment differs from Embodiment 1 in that in step 1, an ethanol solution with a volume concentration of 85% is added for preservation. Other steps and parameters are the same as those in Embodiment 1.

specific Embodiment approach 3

[0037] Embodiment 3: The difference between this embodiment and Embodiment 1 or 2 is that in step 2, stir for 10 minutes and remove large particles of impurities during the stirring. Other steps and parameters are the same as those in Embodiment 1 or Embodiment 2.

Abstract

The invention relates to a method for separating wetland benthic invertebrate rapidly and efficiently, and relates to a separation method of the benthic invertebrate, so as to solve the problems that a conventional manual method for separating benthic invertebrate samples is time-consuming, is large workload and low in efficiency, and easily leads to damages of the benthic invertebrate. The method comprises the steps of acquiring a sample; storing the sample; carrying out flotation by using a NaCl solution, a KCl solution and a sucrose solution; and precipitating to separate the benthic invertebrate. With the method, the damages to the benthic invertebrate produced by conventional hand sorting in batches directly from a mixture of the benthic invertebrate can be prevented. The method guarantees the integrity of the benthic invertebrate individual. Compared with a conventional sorting method directly by the hand, the method provided by the invention reduces separation and purification workload by 2 / 3, shortens time by about 60%, and guarantees the integrity of the benthic invertebrate individual. The method is simple and practical, increases efficiency, guarantees separation accuracy, and has obvious advantages.

Description

technical field [0001] The invention relates to a method for isolating wetland benthic invertebrates. Background technique [0002] Wetland benthic invertebrates refer to aquatic invertebrates that live in the bottom of wetland water bodies during all or at least a period of their life cycle. The benthic invertebrates in the wetland are diverse, including aquatic invertebrates such as aquatic insects, molluscs, oligochaetes, leeches, planarians, molluscs and mites. Benthic invertebrates are important biological components of wetland ecosystems, an important link in the wetland biological food chain, and play an important role in the decomposition of organic matter and water purification. Due to the wide distribution of benthic invertebrates in wetland ecosystems, their sensitivity to environmental changes and their important ecological functions, they are often used as important indicators to evaluate the structural and functional health of wetland systems. Sampling and se...
