The preparation method comprises the following steps of: deodorizing fresh chicken manure, adding sporotrichum thermophile, lactic acid bacteria, thermoactinomyces, brown sugar, crushed straws, flour, corn flour and monopotassium phosphate, fermenting for twice and performing air drying to obtain a preparation I; fermenting bean pulp, wheat bran, calcium powder, brown sugar, bacillus megatherium, bacillus subtilis, lactic acid bacteria and ferrous sulfate for twice, and performing air drying to obtain a preparation II; and mixing the preparation I and the preparation II, adding bacillus megatherium, bacillus subtilis, aspergillus niger, streptomycete, terricolous cyanobacteria, photosynthetic bacteria and dinoflagellate, fermenting, and adding chemical fertilizers needed for growth of the tobacco to obtain the special composite microbial fertilizer for the tobacco.
