Problems solved by technology

[0003] A large number of studies have shown that although Lygus melanogaster is distributed in most parts of China, the external morphological characteristics of this insect are not significantly different among different geographical populations, and it is difficult to distinguish them. Identification is very difficult

Embodiment 1

[0024] Example 1 Method for rapid identification of populations of Lygus clover in Hubei and Gansu provinces and Gansu provinces using mitochondrial molecular markers

[0025] (1) Collect individuals of Lygus clover in Hubei, Hebei, Gansu and Guizhou, and extract whole genome DNA

[0026] Representative populations were selected in the central and border areas of China, that is, individuals of Lygus melanogaster were collected in Hubei and Hebei in the central area and Guizhou and Gansu in the border area. The field collection of Lygus sativa adopts the net capture method, and the adult insects are collected with a suction tube, and then the adults are put into absolute ethanol, and then brought back to the laboratory, and the morphological characteristics of the adults are further identified and confirmed by a body dissecting microscope to ensure Species identification is correct.

[0027] When extracting genomic DNA, the abdomen and wings of adults were removed, and the rem...

Abstract

The invention provides a method for rapid identification of adelphocoris suturalis populations in middle area and edge area by virtue of a mitochondria molecular marker, and in particular identification of populations in Hebei and Hubei and populations in Gansu and Guizhou. According to the method disclosed by the invention, specific primers are respectively designed in accordance with three genes, namely mitochondria cytochrome oxidase I (COI), cytochrome b (Cytb) and mitochondria NADH dehydrogenase 5 (ND5), and PCR amplification products are subject to serial comparison, so that identification of the adelphocoris suturalis populations in middle area and edge area of China is achieved. The method is simple and reliable, strong in operability, good in stability and strong in repeatability.

Description

technical field [0001] The invention relates to the field of molecular biology, in particular to a method for using mitochondrial molecular markers to identify Lygus clover populations. Background technique [0002] As one of the important pest species in cotton fields, Lygus sativa has risen from a secondary pest to a major pest, especially in the past ten years, with the mass planting of transgenic cotton and the reduction in the use of chemical pesticides. The cotton area in the Yangtze River Basin and the Yellow River Basin in my country showed a sharp upward trend. The significant increase in the population of Lygus sativa in a large area in China and the large expansion of its spatial range have brought great challenges to the control work. [0003] A large number of studies have shown that although Lygus melanogaster is distributed in most parts of China, the external morphological characteristics of this insect are not significantly different among different geograph...
