The preparation method comprises the following steps: 1, adding 0.4-0.6 part by weight of the hydrogen-containing silicone oil into 40-45 parts by weight of water; 2, then adding 0.005-0.01 part by weight of octadecyl trimethyl ammonium chloride; 3, then adding 50-55 parts by weight of an evenly-mixed water-mixed gypsum powder, 3-10 parts by weight of the sepiolite powder and 3-5 parts by weight of the montmorillonite powder into the liquid of the step 2; and 4, pouring the mixture into a mold box, forming, demolding, curing with the curing time of 72 h, and thus obtaining the high-strength waterproof gypsum building block.
