Abstract

The invention discloses a QTL-seq method of utilizing / Delta (SNP-index) / for character positioning and application thereof. The method includes that a reference genome is used to replace a genome which is one of parents to serve as a reference to calculate SNP-index of an offspring pool, and an absolute value / Delta (SNP index) / of Delta (SNP index) is used to replace Delta (SNP index) to be used for a sliding window method property positioning analysis; QTL-seq analysis is performed on extreme phenotype pools of segregation populations identical in phenotype of the parents, and distribution of / Delta (SNP-index) / is utilized to perform positioning prediction on a target character gene or a main-effect QTL (quantitative trait loci), so that the circumstance that conventional QTL-seq methods can only be applied in segregation populations with parent homozygosis and contrasting characters is changed, the QTL-seq method is enabled to be applied in segregation populations with parents identical in character, application range of the QTL-seq method is expanded greatly, and the QTL-seq method has good application prospect. The method can be used for analyzing a red-skin phenotype pool and a green-skin phenotype pool of a MantianhongXZaomeisu hybrid combination with red-skin oriental pears to be parents, and oriental pear red-skin / green-skin character sites are positioned within an interval of 1.86 Mb.
