Problems solved by technology

Plant root specimens are different from ordinary plant specimens, which are difficult to collect and cause obvious damage to vegetation, so it is difficult to make
Wheat is a fibrous root, and the collection of fibrous roots is more difficult
Generally, wheat plant specimens only collect a small part of the root system close to the ground. In fact, the root system of wheat is very developed. Limited by the current collection methods and specimen preparation methods, it is particularly difficult to make a complete root system specimen of wheat.
Most of the current wheat plant specimens are dry specimens of mature wheat, and most of them are directly pulled out from the soil, leaving only a few roots, and most of the roots are pulled out, which cannot really reflect the growth status of the wheat roots.
At present, most wheat liquid immersion specimens are only made by selecting the ear and the upper 2-3 nodes of the plant, not the whole wheat plant specimens. This is due to the lack of volume and height suitable for the production of whole wheat plant specimens on the market. Glass containers, for example, the height of glass containers on the market is less than 50cm, while the aboveground part of wheat plants is usually about 80cm, and the plant height of some wheat germplasm resources is more than 1m, so the glass containers on the market cannot satisfy the whole wheat plant. specimen making
So far, there are no soaked specimens of wheat root itself and liquid soaked specimens of whole wheat plants including some above-ground organs.

Embodiment 1

[0040] Embodiment 1 The making of wheat specimen

[0041] At the Jinan Experimental Farm of Shandong Academy of Agricultural Sciences, wheat was planted and sampled, and specimens were made.

[0042] 1. Selection of plants

[0043] In the field plots where wheat is planted, before the wheat turns green, select vigorously growing plants as candidate plants for making specimens, and mark them, and then pull out other unnecessary plant seedlings. Generally, three candidate plants are selected, and the distance between the plants should be kept at 70 cm; or when wheat is sown, the distance between wheat sowing should be kept at 70 cm.

[0044] 2. Selection of earthwork

[0045] During the jointing stage of wheat, around the selected wheat candidate plants, 1m long steel bars with a diameter of 0.5cm are vertically inserted into the soil layer with a radius of 30cm, and the steel bar spacing is about 10cm ( figure 1 ).

Abstract

The invention provides a making method for a liquid-preserved specimen of a wheat plant. The making method comprises the following steps: digging of a wheat plant from field soil; flushing of roots; removal of foreign materials in the roots; chemical treatment of the whole wheat plant; removal of a treatment fluid; soaking with formaldehyde liquid; shaping of a specimen; and hermetic preservation of the specimen in a transparent acrylic synthetic-glass container. The method is applicable to making of liquid-preserved specimens of tissue culture seedlings, whole plants, underground roots or the like of triticeae crops (including transgenic materials) under all soil cultivation conditions and non-soil cultivation conditions.

Description

technical field [0001] The invention relates to a method for preparing a plant specimen, in particular to a method for preparing a specimen soaked in liquid of a wheat plant. Background technique [0002] Wheat is one of the important food crops in my country, and its production level is very important to ensure national food security. Wheat plants are mainly composed of above-ground stems, leaves and spikes and underground roots. The aboveground part is an important place for photosynthesis. The underground part is an important vegetative organ of wheat. The number, weight, thickness, and length of wheat roots directly affect the absorption of water and nutrients by wheat plants, as well as the lodging resistance of individual wheat plants and their populations. Wheat relies on its intricate and massive root system to draw water and nutrients from the soil to feed the wheat's metabolic processes. A strong root system is an important material basis for obtaining high yiel...
