[0145] A total of N=128 subjects with schizophrenia or schizoaffective disorder were included in the study (see Tiwari et al.2010; Tiwari et al.2010; Brandl et al.2012; Brandl et al.2013; Tiwari et al.2013 ). Diagnosis is made according to DSM-III-R or DSM-IV criteria. Subjects were recruited from two different sites: Sample A (N=74; HYM) was collected at Case Western Reserve University, Cleveland, Ohio, USA. The patient was naïve to second-generation antipsychotics and received clozapine for six weeks. Sample B (N=54; JAL) was collected at Hillside Hospital, Glen Oaks, NY, USA. Patients were treated with clozapine, risperidone, olanzapine, or halopirol using a double-blind study design. Written informed consent was obtained from all subjects according to institutional ethical guidelines prior to study entry. Abstract

The invention provides methods and compositions for the treatment of a subject having a psychiatric disease or disorder based upon the subject's genotype and / or the number of risk alleles carried by the subject, which risk alleles have been found by the present inventors to predispose a subject to antipsychotic medication induced weight gain (AIWG). The methods of the invention also provide for different treatments, or different treatment regimens, for the subject depending on the subject's risk of AIWG. Related compositions, in the form of kits, systems, and computer-readable media are also provided.

In particular, the present invention relates to methods and compositions for treating and preventing antipsychotic drug-induced weight gain based on one or more genetic polymorphisms associated with antipsychotic drug-induced weight gain in human subjects .
