Embodiment 1

[0023] Prepare medium with pH5 and pH8, inoculate the mycelium of the overexpressed strain on the medium, wait for about 10 days to count the number of spores produced, and treat the spores of the overexpressed strain with pH5 and pH8 at the same time, observe the spore germination rate and appressoria formation rate, etc. (Table 2).

Abstract

The invention discloses a method for lowering sensitivity of a pyricularia grisea effect protein gene overexpression strain to pH. The method comprises the following steps: treating pyricularia grisea effect protein gene overexpression strain spores with pH5 and pH8; performing statistics to obtain spore yield, sprouting rate and appressorium forming rate. Compared with the prior art, the method has the advantages that the sensitivity of the pyricularia grisea effect protein gene overexpression strain to different pH can be easily, conveniently and accurately determined, and an important reference basis is laid for rapid comprehension of the pathogenicity of a pathogenic bacteria effect protein gene overexpression strain.

Description

technical field [0001] The invention belongs to the field of plant protection and biotechnology, and in particular relates to a method for reducing pH sensitivity of blast fungus effector protein gene overexpression strains. Background technique [0002] Some fungi have evolved complex regulatory mechanisms to recognize and respond to pH changes around them during their persistent interactions with their environment. In addition, some fungi secrete different proteins depending on the pH of their surroundings. In order to successfully invade the host, Botrytis cinerea secretes different proteins in different tissues of the host during the infection process. Although only a few 'true' virulence factors are successful in suppressing host defense responses, different extracellular secreted proteins are required to act in concert. Seven proteins in Aspergillus nidulans are related to pH recognition ( et al., 2008), two of the proteins, PalH and Pail, are putative membrane pro...
