Abstract

The invention provides a method for studying differentially expressed proteome in brain tissues of mice chronically infected with toxoplasma gondii by an iTRAQ (Isobaric Tags For Relative And Absolute Quantitation) technology. The invention also provides a protein or polypeptide interacting with developmentally regulated brain protein and application of the protein or polypeptide. In the invention, six toxoplasma gondii proteins interacting with the developmentally regulated brain protein are found by CoIP and Pulldown experiments; and gene knockout experiments and neurobehavioral validation are further performed on TgRPS14 gene and TgH4 gene. The method for studying differentially expressed proteome in brain tissues of mice chronically infected with toxoplasma gondii provided by the invention can rapidly and effectively perform proteomics research, and is beneficial for rapidly positioning to detection targets and prevention and treatment medicine target points with a research value or a commercial application value.
