Abstract

The invention discloses a celosia cristata explant culture method, which includes the following steps: (1) celosia cristata seeds are inoculated into germination medium; (2) the germination medium inoculated in step (1) is cultured under the temperature of 25 DEG C to 28 DEG C, the illumination intensity of 1200lx to 1500lx and the illumination time of 14h / d for 10 days; (3) the hypocotyledonary axes of aseptic celosia cristata seedlings obtained in step (2) are cut into small explant sections which are 0.8mm to 1.2mm on average, the small explant sections are then inoculated into inducing medium, and the inducing medium is added with compound growth regulator compounded from 2,4-D, the concentration of which is 0.1mg / L, and BA, the concentration of which is 0.5mg / L; (4) the inducing medium in step (3) is cultured under the same culture conditions as step (2) for 10 days. Since the optimal culture conditions from celosia cristata seed germination to explant callus induction in the process of culture are determined by the method, the inductivity of the cultured celosia cristata explants is high, the growth of calluses is fast, and moreover, the content of callus anthocyanin is high. The method is applicable to the mass production of celosia cristata calluses and anthocyanin.

Description

technical field [0001] The invention relates to the field of plant cultivation, in particular to an explant cultivation method. Background technique [0002] Anthocyanins, also called anthocyanins, are flavonoids. As the most famous natural pigment, it widely exists in the plant kingdom and is a water-soluble pigment in plants. Due to the different molecular structures or physicochemical properties of each anthocyanin, it presents different colors, and when combined with different metal ions, it forms colorful coordination compounds, which are pleasing to the eye. However, due to the properties of natural pigments and the superiority of synthetic pigments in terms of color, stability, tinting strength, price, etc., after the 1950s, the use of natural pigments gradually decreased, while the use of synthetic pigments increased. However, synthetic pigments not only have no nutritional value, but in the past two decades, toxicology and biological tests have confirmed that synt...
