Embodiment 1

[0040] Embodiment 1, the grafting method of peanut cultivar tissue culture seedling

[0041] A method for grafting peanut tissue culture seedlings, comprising the following steps:

[0042] (1) Preparation of rootstock material: choose mature and plump black peanut Yuhua 0215 seeds, soak them overnight to accelerate germination, plant the seeds in a nutrient bowl (autoclaved vermiculite) after dew whitening, then pour water thoroughly, and place them in the dark at 27°C Cultivate, when the two true leaves of peanut seedlings unfold (about 7d) (see figure 1 ), select the seedlings whose hypocotyl length is 6-8cm as rootstock material.

[0043] (2) Preparation of sterile tissue-cultured seedling scions: Transgenic resistant seedlings of Yuhua 22, Yuanza 9307 and Yuanza 9102 were used as scions respectively, and grafted (the height of the tissue-cultured seedlings was more than 2 cm).

[0044] (3) Grafting: cut off the cotyledons and the above parts of the peanut seedlings (root...

Embodiment 2

[0051] Embodiment 2, the grafting method of peanut wild species tissue culture seedling

[0052] A method for grafting peanut tissue culture seedlings, comprising the following steps:

[0053] (1) Preparation of rootstock material: choose mature and plump black peanut Yuhua 0215 seeds, soak them overnight to accelerate germination, plant the seeds in a nutrient bowl (autoclaved vermiculite) after dew whitening, then pour water thoroughly, and place them in the dark at 27°C Cultivate, when the two true leaves of peanut seedlings unfold (about 7d), select the seedlings whose hypocotyl length is 6-8cm as rootstock material.

[0054] (2) Preparation of aseptic tissue-cultured seedling scions: use wild species (A.cor, A.dard, 30119) and progeny materials (ampiploid Am-L) as scions respectively, and carry out grafting (tissue-cultured seedlings are at a height of 2cm or more).

[0055] (3) Grafting: cut off the cotyledons of peanut seedlings (rootstock) and the above parts, longit...

Abstract

The invention discloses a grafting method of a peanut tissue-cultured seedling. The grafting method comprises the following steps of culturing a black-peanut seed in a dark culture condition, when twotrue leaves of a peanut seedling spreads, selecting the seedling whose hypocotyledonary axis length is 6cm to 8cm as a stock, cutting off a seed leaf of the seedling and a part above the seed leaf, and longitudinally cutting the middle of a hypocotyledonary axis into a notch; using an aseptic tissue-cultured seedling as a scion, grafting and transplanting into a large field. According to the grafting method, the seedling is cultured by utilizing a dark condition, the hypocotyledonary axis of the seedling can be enabled to quickly grow; the seedling is used as the stock to begin grafting whenthe hypocotyledonary axis length is 6cm to 8cm; the problems that the hypocotyledonary axis of a stock material which is subjected to ordinary illumination culture is shorter (1cm to 2cm) and the operation is difficult during the grafting can be avoided; the grafting efficiency and the grafting survival rate are directly improved. Further, in comparison with the situation that a dozen seedlings can be only grafted within 1h when the hypocotyledonary axis is shorter, the grafting method can be used for grafting 30 seedlings or more within 1h when the hypocotyledonary axis is longer and the workefficiency is increased by more than one time.

Description

technical field [0001] The invention belongs to the field of agricultural planting biotechnology, and in particular relates to a method for grafting peanut tissue culture seedlings. Background technique [0002] Peanut is an important oil crop and economic crop, and occupies an important position in agricultural production. In recent years, the peanut planting area in my country has gradually expanded. Affect the further improvement of the yield and quality of peanuts. With the rapid development of biotechnology, transgenic and in vitro mutagenesis techniques have attracted the attention of many researchers; with the gradual improvement of peanut distant hybridization technology, wild species with excellent traits such as disease resistance and stress resistance are selected for hybridization with cultivated species. It has become an important way to breed new varieties with good disease resistance and stress tolerance. However, most distant hybrids show hybrid incompatibili...
