Problems solved by technology

[0003] Meat samples have a complex matrix and high fat content. Traditional pretreatment methods include solid phase extraction (SPE) and gel permeation chromatography (GPC). However, the cost of SPE is high, and GPC requires a large amount of organic solvents for elution.

Embodiment 1

[0076] The method for extracting environmental hormone compounds from fish meat is to investigate the influence of different centrifugation speeds on the extraction efficiency, wherein the centrifugation speeds are 4000rpm, 6000rpm, 8000rpm, 10000rpm, and other steps and conditions are the same as steps 1-7. The standard recovery and relative standard deviation (RSD) of environmental hormones in the organic extractant after extraction were investigated. See Table 5 for the results of the investigation.

[0078]

[0079]

[0080] It is known from Table 5 that the recovery rate increases with the increase of the rotational speed, indicating that the extraction effect is positively correlated with the centrifugal speed. At a rotational speed of 4000-6000rpm, the recovery rate is low, and when the rotational speed increases from 8000 to 10000, the recovery ra...

Embodiment 2

[0082] For the analysis of different types of freshwater fish caught in the Changsha Basin of the Xiangjiang River, the analysis steps include:

[0083] Wild silver carp, bream and catfish in the Xiangjiang River are used as three common freshwater fish in South China. Silver carp mainly inhabits the middle and upper layers, bream mainly inhabits the middle layer, and catfish is a bottom-dwelling fish. Homogenize the fish tissue, weigh 5g of minced fish in a centrifuge tube, and other steps and conditions are the same as steps 1-7. To investigate the detection content and recovery rate of environmental hormones in different kinds of fish meat.

Abstract

The invention discloses a determination method for environmental hormones in a meat matrix sample. The method includes the steps of: placing a minced meat sample in a plug equipped centrifuge tube, adding an internal standard substance, acetonitrile and an inorganic salt, conducting ultrasonic treatment and centrifugation to obtain an acetonitrile extracted liquid; taking the acetonitrile extracted liquid and putting it to a slender-neck extraction bottle, adding purification filler and ultrapure water, an inorganic salt and an organic extractant; using a glass syringe to suck the solution inthe extraction bottle, then injecting the sucked solution into the extraction bottle, repeating suction and injection to mix the solution evenly, wrapping the extraction bottle with plasticine in thecentrifuge tube and performing centrifugation to obtain a three-phase separated clarified solution; inserting an ultrapure water loaded long needle glass syringe into the bottom water phase, injectingultrapure water to make an upper organic phase float and rise to the slender neck of the extraction bottle; and using a microsyringe to suck the organic phase at the slender neck of the extraction bottle as a to-be-tested sample, testing the to-be-tested sample with a liquid chromatogram-mass spectrometer, and calculating the concentration of environmental hormones in the minced meat sample.

Description

technical field [0001] This patent relates to the technical field of food testing and environmental monitoring, in particular to a sample pretreatment method and detection method for extracting the content of environmental hormones from meat matrix. Background technique [0002] Environmental hormones refer to the ability to stabilize the internal environment of the body by interfering with the normal activities of synthesis, secretion, transport, binding, metabolism, and clearance of hormones in humans and animals, or by inhibiting the functions of the endocrine system. Exogenous substances that affect human body, metabolism, development, reproduction, etc. Nearly 20 common environmental hormones (including 4 progestogens, 6 androgens, 4 estrogens and 6 alkylphenols) are the main research target substances. More and more environmental hormones enter water bodies through atmospheric deposition, surface runoff, soil leaching, and direct discharge. Water resources and freshwa...
