Embodiment Construction

[0025] The principles and features of the present invention are described below in conjunction with examples, which are only used to explain the present invention and are not intended to limit the scope of the present invention.

[0026] 1. Sample collection and grouping

[0027] All testicular tissue samples were obtained from the Center for Reproductive Medicine, Tongji Medical College, Huazhong University of Science and Technology, and the research was approved by the Ethics Committee and informed consent was obtained from the patients. Micro-TESE operation finds the source of sperm group samples is excess discarded testicular tissue after NOA patients have given birth to normal offspring through assisted reproductive technology in this reproductive medicine center, and the source of no sperm group samples is after micro-TESE surgery in this reproductive medicine center Discarded testicular tissue where no sperm was found. According to whether micro-TESE obtained sperm, th...

Abstract

The invention relates to application of piRNA for preparing a reagent for diagnosing whether sperm remains inside the testis of an NOA patient or not, and further relates to the reagent and a kit fordiagnosing whether sperm remains inside the testis of the NOA patient or not. The piRNA is adopted as a molecule marker, whether sperm exists inside the testis of the NOA patient is diagnosed by detecting the expression level, and therefore the surgery sperm extraction success rate of the NOA patient can be judged through micro-TESE and other methods in advance, unnecessary surgery and testis wounds of about 60% of NOA patients can be avoided, and great social and economic benefits are achieved.

Description

technical field [0001] The present invention relates to the field of human reproductive health, more particularly, relates to the application of miRNA in the preparation of reagents for diagnosing whether there is residual sperm in the testis of NOA patients, and also relates to the reagent and kit for diagnosing whether there is residual sperm in the testis of NOA patients. Background technique [0002] Infertility is a common reproductive disease with an incidence of about 15% among couples of childbearing age. Among male infertility, the most serious type is azoospermia, accounting for about 1% of the male population of reproductive age. Among all the causes of male infertility, spermatogenesis disorder is the most common factor, and it is also one of the most important threats to the reproductive health of adult males. Impairment of spermatogenesis is a multi-factor and multi-stage process. The main clinical manifestation is the decrease of sperm density in ejaculated s...
