Abstract

The response surface optimization method for extracting barley polyphenols belongs to the technical field of barley polyphenol extraction. The invention adopts an ultrasonic method to extract coix seed polyphenols, uses gallic acid as a standard product and folinol as a color developing agent, and measures the polyphenol content by a colorimetric method. The experiment investigated the effects of seven factors including solvent type, solid-liquid ratio, solvent volume fraction, extraction temperature, extraction time, extraction times, and sample particle size on the extraction rate of barley polyphenols. On the basis of the single factor experiment, the experimental scheme was designed by using the Box-Behnken response surface analysis method, and the material-liquid ratio, solvent volume fraction, extraction time, and extraction temperature were used as the experimental factors. The effectiveness of the regression model and the influence of each single factor were analyzed, and the factors in the extraction of polyphenols from the seed of Job's tears were optimized to obtain the optimized process conditions for the extraction of polyphenols from the seed of Job's tears. There is a good fit between the measured value and the theoretical value, which is suitable for the extraction of polyphenols from barley.
