Abstract

The invention discloses a method for rapid propagation of embryogenic cells of fraxinus mandshurica. The method comprises the following steps of selecting calluses of fraxinus mandshurica, wherein thecalluses can be originated from various explants such as roots, stems, leaves, seeds and embryos, and inoculating the calluses to an F4 solid medium to cultivate 1-2 subcultures so as to obtain embryogenic calluses of fraxinus mandshurica; and inoculating friable embryogenic calluses to an F4 liquid medium to cultivate 2-3 subcultures so as to obtain single cells and small cell groups of fraxinusmandshurica. The method has the advantages that the F4 media are selected; besides conventional WPM basic media, 30g / L saccharose is selected to be combined with four hormones: BA, NAA, IBA and TDZ;cells on the media grow vigorously, and no death is caused by browning as long as the subculturing takes place in the growth cycle. The method is established to lay a foundation for scaled productionof cells of fraxinus mandshurica and researches on secondary metabolites.
