Abstract

The invention belongs to the technical field of medical models, and discloses an animal heart failure model and a construction method thereof. The construction method of the animal heart failure modelcomprises the following steps: obtaining an 8-week-old SPF-grade male C57/BL6 mouse, putting the mouse into a purification cage, controlling the temperature at 19-23 DEG C and the relative humidity at 40-50%, keeping bright illumination and dark illumination, each for 12 h, feeding the mouse with a standard mouse feed, and allowing freely drinking of water so as to carry out adaptive feeding; after 1 week of the adaptive feeding, implanting the mouse with a microosmotic pump containing oxidized low density lipoprotein, and waiting for 4 weeks so as to obtain the animal heart failure model; and then, determining degree of heart failure by adopting echocardiography, plasma BNP level and the like. Different from previous heart failure models induced by pressure load, volume load, myocardialischemia and cardiotoxic drugs, the animal heart failure model prepared by the construction method fills in the blank of animal models with heart failure of unknown causes, and pertains to novel animal heart failure models with the heart failure caused by hyperlipidemia. The animal heart failure model is good in repeatability, simple, and objective.

Description

technical field [0001] The invention belongs to the technical field of medical models, and relates to an animal heart failure model, a construction method and application thereof, in particular to a construction method of an obesity-related heart failure animal model. Background technique [0002] Heart failure is the end stage of heart disease in which the reduced ventricular pumping function cannot meet the needs of the body's life activities. It is a common clinical cardiovascular syndrome, and its morbidity and mortality are constantly increasing. The etiology of heart failure is complex, such as myocardial ischemia, increased cardiac pressure load, increased cardiac volume load, excessive activation of neuroendocrine cytokines, primary cardiomyopathy and other causes can cause ventricular remodeling and lead to heart failure symptoms. It is of great significance to identify new therapeutic targets for heart failure and develop new preventive measures. Animal experiments...
