Problems solved by technology

[0006] Using the traditional method to process the seeds of Orchid grandiflorum, the seeds are difficult to germinate, the germination rate is low, and the germination cycle is long, but there are few methods in the prior art that can germinate the seeds of Orchid grandiflora in advance.

Embodiment 1

[0025] (1) Harvesting and pretreatment of seeds: After the pods of Orchid grandiflorum turn yellow and mature, they are collected back to the laboratory, dried a little naturally, cut open the pods with a scalpel, carefully take out the seeds, and put them into a weigher. In a seed drying bag made of measuring paper, place the drying bag in a sealed drying dish and slowly dehydrate and dry it with lithium chloride, then store the seeds in an Asone high borosilicate glass sealed sample bottle, and then place the sample bottle in a Store in a silicone sealed box for later use;

[0026] (2) Seed sterility treatment: make a seed bag with weighing paper, take out a small amount of Orchid grandiflora seeds from the sealed sample bottle and put it into the seed bag, seal the seed bag, and put it on the ultra-clean workbench , placed in a beaker with 75% alcohol, sterilized for 5-10s, rinsed with sterile water for 3-5 times, then sterilized in 10% NaCLO solution for 7min, and rinsed w...

Abstract

The invention discloses a method for promoting germination of pleione grandiflora seeds. The method comprises the following steps of (1) seed harvesting and pretreatment, specifically, after pleione grandiflora capsules become yellow and mature, harvesting the pleione grandiflora capsules back to a laboratory, naturally and slightly drying the pleione grandiflora capsules, taking out the seeds, drying the seeds, and storing the seeds for later use; (2) carrying out aseptic treatment on the seeds; (3) carrying out sterile seeding treatment, specifically, sucking seed solutions by using a disposable large-size injector, uniformly transferring the seed solutions to a sterile germination culture medium with the pH value of 5.8 as little as possible, and slightly shaking a culture dish to uniformly disperse the seeds; and (4) after inoculation is finished, putting the culture dish under the conditions that the temperature is 25(+ / -1) DEG C, the illumination intensity is 800-1200 lx and theillumination is 12h / d for culturing. By adopting the method for treating the pleione grandiflora seeds, the time required for germination of the pleione grandiflora seeds can be greatly shortened, andmeanwhile the germination rate of the pleione grandiflora seeds can be remarkably increased.

Description

technical field [0001] The invention relates to the technical field of orchidaceae plant propagation, in particular to a method for promoting the germination of orchid orchid seeds. Background technique [0002] Orchidaceae (Orchidaceae) belongs to the class Monocots of the angiosperm phylum, and there are about 700 genera and more than 35,000 species distributed in the world (Dressler, 1993; Cribb, 2003), and its number of species ranks second only to Compositae. Branch (Lang Kaiyong, 1994). It is beautiful in shape, bright in color, and has great ornamental and medicinal value. [0003] Pleione grandiflora Rolfe is a rock-growing or epiphytic herb of the Sect.Humiles group of the genus Pleione. It mostly grows in the understory or on the edge of the forest and is covered with moss. Some grow on rocks, and some grow on moss-covered tree trunks. This species is a species with high ornamental value in the genus Sulicia, and is suitable for small potted flowers. Its flower ...
