The preparation method comprises the following steps: (1), preparing a duck reovirus antigen and a duck hepatitis virus liquid antigen; (2), inactivating the prepared antigen, adding an oil adjuvant, performing mixing and emulsifying to obtain a duck reovirus-caused disease and a duck viral hepatitis bivalent inactivated vaccine; and (3), immunizing the laying hens by using the bivalent inactivated vaccine, collecting high-immunity eggs, and extracting egg yolk antibodies. According to the invention, the antibody immune challenge test result is visible; the antibody provided by the invention can safely and effectively prevent novel duck reovirus-caused disease with liver irregular necrosis and spleen necrosis as characteristic lesions and duck viral hepatitis with liver swelling and bleeding as main pathological changes caused by duck hepatitis virus; and two epidemic diseases of the novel duck reovirus disease and the duck viral hepatitis can be prevented and treated by one injection.
