Problems solved by technology

At present, clinical treatment selection and prognosis evaluation of liver cancer are mainly based on liver cancer grading and staging evaluation systems such as TNM staging, BCLC staging, ALBI grading, etc., but the pathological characteristics at the molecular level are not considered, and the guiding role is limited.
However, studies on molecular classification mainly focus on genomics and proteomics. A large number of serum protein markers, miRNAs, and LncRNAs have been found to be closely related to the prognosis of patients with liver cancer, which also provides a certain basis for the explanation of the pathogenic mechanism. Effects are still limited, so more attempts at prognostic risk stratification at the molecular level in HCC are warranted
At present, there is no report on the combination of the above two metabolites and clinical features-the presence or absence of sub-foci next to the cancer tissue to evaluate the prognosis risk of liver cancer

Embodiment 1

[0030] 1 Collection of serum samples: 78 patients with liver cancer in Dongfang Hepatobiliary Surgery Hospital were included in this study from June 2012 to July 2012, starting from the operation time, and followed up for 5 years after operation. The serum samples of the subjects were collected under the same conditions before the operation, and stored in a -80°C refrigerator before metabolomics analysis. All subjects signed a written consent form before sample collection. The study protocol was approved by the Ethics Committee of the Eastern Hepatobiliary Surgery Hospital and performed in accordance with the guidelines of the 1964 Declaration of Helsinki, as revised in 1975.

Abstract

The invention relates to a kit prepared by using a combination of small molecule metabolites phenylalanine and choline in a serum sample and clinical pathological characteristics (whether sub-lesions exist beside cancer tissues) as a combined marker, and a new application of the kit in the prognosis risk of liver cancer of a subject. Three variables including relative concentrations of the two metabolites in a serum sample of the subject and clinical characteristics (whether sub-lesions exist beside cancer tissues) are detected, a prognosis risk score of the liver cancer patient is calculated on the basis of a Cox risk proportion regression equation, and a node value is judged so as to evaluate the total survival time of the liver cancer patient; it is found that the total survival time of patients in a high-risk group is significantly shortened compared to patients in a low-risk group. In addition, compared with a traditional liver cancer clinical grading system, the system has the advantages of being rapid, accurate and low in detection cost, and can be used for assisting and guiding prognosis risk layering and individualized treatment of clinical liver cancer patients.

Description

technical field [0001] The invention relates to the application of small molecular metabolites phenylalanine and choline in serum samples and the presence or absence of subfoci next to cancer tissue as risk factors in the preparation of a kit for evaluating the prognosis risk of liver cancer. It belongs to the fields of analytical chemistry, clinical medicine and medicine. Background technique [0002] Liver cancer (HepatocellμLar carcinoma, HCC) is one of the tumors with the highest morbidity and mortality in the world, and East Asia, especially my country, is the hardest hit area for liver cancer. In recent years, with the advancement of liver cancer diagnosis and treatment methods, surgery, TACE, radiotherapy, chemotherapy, and new targeted drug therapy have provided more options for liver cancer treatment. The comprehensive application of these methods has improved the prognosis of liver cancer to a certain extent. Among them, surgical treatment is still the main means ...
