Embodiment 1

[0073] In the 10mM chloroplatinic acid solution of 10mL, add 111.1mg of polyvinylpyrrolidone (PVP), after stirring for 15 minutes, after adding the 100mM sodium borohydride solution of 200uL newly configured therein, stir for 12 hours, the solution becomes Dark brown, platinum nanoparticles were obtained after filtration and washing, and then the platinum nanoparticles were mixed with an appropriate amount of water to prepare an aqueous solution of platinum nanoparticles with a mass concentration of 1 mg / mL.

[0074] Add 2-methylimidazole (0.26g, 3.15mmol) and the above platinum nanoparticle aqueous solution (0.1mL) into 0.9mL of water and mix evenly, then add 100 μL of 0.5M zinc nitrate and nitric oxide synthase (0.6 mg) mixed aqueous solution (the molar number of zinc nitrate is 0.05mmol), after the reaction, the cascade nanozyme is obtained by centrifugal filtration and washing, which is denoted as ZIF@Pt@NOS, and its TEM image is as follows figure 2 As shown, the particle...

Abstract

The invention discloses a cascaded nano-enzyme as well as a preparation method and application thereof. The cascaded nano-enzyme comprises a metal organic framework carrier, nitric oxide synthetase combined with the metal organic framework carrier and platinum nanoparticles combined with the metal organic framework carrier. The cascaded nano-enzyme belongs to a nano size, has the natural advantage of passive accumulation in the liver compared with a small molecule drug, can increase the enrichment of the drug in the liver, and has higher targeting property. The platinum nanoparticles as the nano-enzyme have excellent reactive oxygen species (ROS) scavenging capacity and can catalyze the reactive oxygen to generate oxygen, the generated oxygen reacts with arginine under the catalysis of nitric oxide synthetase (iNOS) to generate NO in situ, and the bioavailability of NO is improved. Therefore, the cascaded nano-enzyme improves the targeting property and bioavailability of NO, and can simultaneously realize ROS scavenging and NO regulation, thereby realizing effective intervention on injury caused by liver ischemia reperfusion.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to a cascade nanozyme and its preparation method and application. Background technique [0002] The rapid development of liver surgery is inseparable from the progress of surgical anatomy, but also benefited from the application of new materials and technologies. Liver resection and liver transplantation are the most common methods for clinical treatment of liver diseases such as benign and malignant liver tumors, intrahepatic bile duct stones, and liver trauma. Liver surgery usually needs to block the hepatic hilum to control blood flow and reduce blood loss. The liver will be partially damaged during ischemia. When the blood flow is restored, the damage to the hypoxic organ will be aggravated, resulting in hepatic ischemia-reperfusion injury (Hepatic IschemiaReperfusion Injury) , HIRI). When HIRI occurs, a large amount of reactive oxygen species (ROS) and cellular inflammatory factors...
