Embodiment 1

[0038] 1. Microarray data analysis

[0039] Use the GEO database to screen eligible research series: search for postmenopausal osteoporosis-related gene expression microarray results from the GEO database http: / / www.ncbi.nlm.nih.gov / geo / , and the search criteria are limited to: ( 1) Postmenopausal osteoporosis; (2) The species is of human origin; (3) The research type is microarray expression profiling; (4) The source of the specimen is peripheral blood samples. After screening, two microarray series were included in the research: GSE56116 and GSE100609.

[0040] In the present invention, there are 13 peripheral blood samples in GSE56116, 3 of which are postmenopausal healthy women as the control group, and 10 postmenopausal osteoporosis patients are the experimental group; there are 8 peripheral blood samples in GSE100609, of which 4 A healthy postmenopausal woman served as the control group, and 4 postmenopausal osteoporosis patients served as the experimental group.

Abstract

The invention discloses a postmenopausal osteoporosis related gene screening and function analysis method, which comprises the following steps: searching a gene expression chip related to postmenopausal osteoporosis from a GEO database, and obtaining a gene expression result; searching overlapped differential genes in the data sets of the two chips by utilizing a Venn graph; and carrying out gene enrichment function analysis by using a Metascap database and a DAVID database. According to the method, expression differential genes are downloaded by utilizing multiple online databases, the differential genes jointly expressed in two chip data sets are searched, bioinformatics analysis is carried out on the jointly expressed differential genes, and meaningful exploration and basis are provided for marker screening, molecular pathogenesis and the like related to the postmenopausal osteoporosis.

Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to a method for screening and functional analysis of genes related to postmenopausal osteoporosis. Background technique [0002] Osteoporosis (Osteoporosis, OP) is a systemic skeletal disease characterized by decreased bone mineral density (BMD) and degeneration of bone microstructure, resulting in increased bone fragility and susceptibility to fracture. Postmenopausal osteoporosis (postmenopausal osteoporosis, PMOP) is the osteoporosis caused by ovarian function decline, estrogen level decline, and bone loss increase in postmenopausal women. The most serious consequence of osteoporosis is fracture, and there are approximately 9 million clinical osteoporotic fractures worldwide each year. Among women over 60 years old in my country, the incidence of osteoporosis is as high as 40-50%, of which 30-50% will experience osteoporotic fractures, and osteoporotic fractures have a higher morbidit...
