Problems solved by technology

[0006] Despite various methods of expanding chondrocytes and verifying their quality, sufficient numbers of phenotypically stable chondrocytes cannot be obtained in a defined number of passages to improve the characteristics of low-quality chondrocytes or to recover after inappropriate handling (eg, excessive Incubation or expansion) recovery of cell properties

Embodiment approach

[0085] According to a specific embodiment of the invention, the regulatory cell population and the chondroblast population used in the combined cell population of the invention are obtained from the same cartilage sample. According to a first embodiment, a portion of the freshly isolated cells obtained from the biopsy sample is frozen, either directly or within 24 hours of the first expansion, and the second portion is expanded to obtain an expanded chondrocyte population (population of mature chondrocytes). Alternatively, both the regulatory cell population and the chondrocyte population are derived from the same first expansion of a freshly isolated chondrocyte sample, where the regulatory cells correspond to the cell population taken from the non-adherent cell fraction during the first expansion of chondrocytes, Mature chondrocytes correspond to one or more chondrocyte passages of the first expanded adherent cell population of the same chondrocytes. Alternatively, it is en...

Abstract

The present invention relates to regulatory cells, which are capable of restoring, maintaining or improving the stable cartilage phenotype of expanded and passaged chondrocytes. These regulatory cells are also capable of directing precursor and stem cells into the chondrogenic lineage. An enriched population of regulatory cells can be obtained by harvesting the non-adherent cells in the culture medium of a monolayer culture of PO chondrocytes.

Description

technical field [0001] The present invention relates to the field of chondrocyte expansion and cartilage repair. The present invention also relates to controllable cell populations of cartilage that can be used to maintain, improve or restore the cartilage phenotype of chondroblasts such as chondrocytes or chondroblasts. Background of the Invention [0002] Both articular cartilage and meniscus cartilage play important roles in normal joint movement mechanisms. Articular cartilage covers the ends of the bones of the knee joint. It consists of sparsely distributed cells (chondrocytes) enclosed in an extracellular matrix consisting of collagen, various proteins / glycoproteins, and highly water-retaining proteoglycans carrying negatively charged polysaccharides called mucopolysaccharides . Meniscal cartilage is fibrocartilage characterized by the appearance of cells in lacunae arranged in rows and bundles of collagen fibers in the matrix. Injury to articular cartilage and / or...
