The preparation method comprises steps of: weighing wood chip, cotton straw, wheat bran, rice bran, corn flour, cob and cottonseed casing according to the proportion and mixing; dumping a mixture into a pulverizer and crushing the mixture into particles about 120 mesh; adding gypsum powder according to the proportion, stirring in a mixer for 65 min; supplementing a proper amount of water till a water content of 64-66%; and disinfecting to realize a pH of 5.5-7.5. The invention has beneficial effect that because of employment of the above formula, the prepared culture material for hypsizigus marmoreus has low cost, low pollution rate, fast spawn running effect, and a single bottle mushroom yield similar to that of a culture material using cottonseed casing and cob, and high comprehensive benefit.
