Embodiment Construction

[0022] The present invention will be further described in detail below with reference to specific samples, which are to explain rather than limit the present invention.

[0023] 1. Population survey and sampling

[0024] Spartina alterniflora in Luoyuan Bay, Fujian is the most primitive population in my country, and it was used as the sampling point in the area where invasive alien species was first introduced. Shanghai Chongming Island is relatively close to the analysis point, and the samples are highly preserved after collection, so it is used as the sampling point.

[0025] Select well-grown, pest-free leaves, put them into a pre-prepared ziplock bag containing about half a bag of silica gel immediately after harvesting, and mix well to fully mix the leaves and silica gel for rapid dehydration. After taking it back to the laboratory, replace the discolored silica gel according to the actual situation. After drying, it will be used for DNA extraction. About 30 plants are ...

Abstract

The invention relates to a DNA molecular marker method for identification of the alien invasive species Spartina alterniflora population. The method includes: conducting high-throughput transcriptome sequencing and screening to obtain a target gene, carrying out a PCR reaction, subjecting the PCR product to sequencing so as to obtain a plurality of single nucleotide polymorphism (SNP) sites, then letting all effective SNP sites compose a haplotype for molecular markers, thus achieving genetic typing. According to the method provided in the invention, rich polymorphism information can be obtained in one experiment. SNP markers have genetic stability, can be used for samples of different sources, and are suitable for high-throughput automated analysis.

Description

technical field [0001] The invention belongs to the technical field of ecological engineering, and relates to a DNA molecular marker method for identifying populations of alien invasive species Spartina alterniflora. Background technique [0002] Alien biological invasion has become one of the most urgent problems in environmental protection today, and it has a very important impact on the economy, ecology, social security and international trade of all countries in the world. The direct and indirect cost of invasive species management in the United States alone amounts to $137 billion a year. China has also become one of the countries most seriously affected by the invasion of alien species in the world. 488 species of invasive alien species have been discovered, of which more than 50 are among the 100 most threatened alien species in the world announced by the International Union for Conservation of Nature. The direct economic loss has reached 120 billion yuan, and the in...
