Embodiment 1

[0012] How to make a color chart

[0013] (1) Preparation of standard solution: use 0.8 mg / mL HMF standard stock solution, and prepare concentrations of 0, 3, 6, 9, 12, 15, 18 μg / mL (equivalent to the content of HMF in honey is 0, 10 , 20, 30, 40, 50, 60 mg / kg) standard solution;

[0014] (2) Color development: Take 1.00 mL of each of the above standard solutions in a stoppered graduated test tube, then add 3 mL of the mixed solution to the test tube, shake and mix immediately, and develop color for 5 minutes. According to the obtained color, a color chart is made, and the content range of HMF corresponding to the color chart is 0-60 mg / kg.

[0015] Actual sample measurement

[0016] (1) Preparation of samples: Accurately weigh 7.5g of wild osmanthus honey into a small beaker, add 10mL of distilled water to mix, stir well with a glass rod, transfer to a 25mL volumetric flask, add distilled water to volume, shake uniform;

Embodiment 2

[0020] The making method of color card (same as example 1)

[0021] Actual sample measurement

[0022] (1) Sample preparation: Accurately weigh 7.5g of loquat honey into a small beaker, add 10mL of distilled water and mix well, stir well with a glass rod, transfer to a 25mL volumetric flask, add distilled water to the mark, and shake well ;

[0023] (2) Color development: Take 1.00 mL of the sample to be tested in a stoppered scale test tube, then add 3 mL of the mixed solution into the test tube, shake and mix immediately, and develop color for 5 minutes;

[0024] (3) Detection: By comparing the color of the solution after color development with the color of the color comparison card, the color is between the color of HMF content of 40 mg / kg and 50 mg / kg, so the hydroxymethylfurfural in the honey sample is determined The content is 45 mg / kg.

Abstract

The invention discloses a rapid detecting method for hydroxymethyl furfural (HMF) in honey. The rapid detecting method comprises the following steps: (1) preparing a sample, namely accurately weighing and adding 7.5g of honey sample into a small beaker, adding 10mL of distilled water, uniformly mixing the distilled water and the honey sample by a glass rod to obtain a mixture, transferring the mixture to a 25mL volumetric flask, adding distilled water to make up to volume, and uniformly shaking; (2) developing color, namely taking and adding 1.00ml of a to-be-detected sample into a graduated test tube with a stopper, then, adding 3mL of mixed solution into the test tube, and developing color for 5 minutes after uniformly mixing; (3) detecting, namely comparing the color of a developed solution with a color chart to determine the content of HMF. The rapid detecting method for hydroxymethyl furfural in honey disclosed by the invention has characteristics of a simple used reagent, no complex condition and apparatus equipment, simplicity and convenience in operation and rapidness.

Description

technical field [0001] The invention specifically relates to a rapid detection method for hydroxymethylfurfural in honey, which belongs to the technical field of food quality and safety detection. Background technique [0002] Hydroxymethylfurfural (HMF) is absent in fresh honey, and is converted from fructose during storage or heating of honey, so it is one of the important signs to distinguish whether honey is fresh. According to international regulations, hydroxymethylfurfural in honey should not exceed 40mg / kg. At present, there are three main methods for determining the HMF content in honey: chemical chromogenic method, high performance liquid chromatography, and ultraviolet spectrophotometry. Although these methods are relatively accurate, the experimental steps are complicated, time-consuming, and require related instruments and equipment, so they are not suitable for on-site rapid detection. Therefore, on the basis of predecessors' research on hydroxymethylfurfural...
