Problems solved by technology

Currently known tuberculosis CTL epitope peptides are mainly identified in European and American populations, most of which are HLA-A*0201-restricted, but for African and Southeast Asian populations where tuberculosis is most prevalent, there is no suitable epitope peptide to use

Embodiment Construction

[0016] According to the primary structure of the antigen, the present invention uses immunoinformatics means to predict and analyze the HLA-A*1101-restricted CTL epitope peptide of the Mycobacterium tuberculosis antigen protein by using the NetMHCcons epitope prediction software in the CBS database, and then The affinity and stability of candidate peptides binding to HLA-A*1101 molecules were tested by UV-induced peptide displacement experiments, and epitope peptides were obtained by screening. Through in vitro experiments, CTL epitope peptide P12 can induce CD8 + T cells generate a strong cellular immune response, secrete high levels of IFN-γ, and induce CD8 + T cells proliferate significantly.

[0017] The content of the present invention will be further described below in conjunction with specific examples.

Abstract

The invention discloses a mycobacterium tuberculosis specific CD8<+>T cell epitope peptide and an application thereof. The amino acid sequence of the epitope peptide is AVMAGIVRAAK. The epitope peptide has high affinity with a molecule HLA-A*1101 and can be used for activating a CD8<+>T cell and inducing the CD8<+>T cell to proliferate and secrete IFN-gamma. Determination of the epitope peptide provides a theoretical basis and a new solution for research and development of tuberculosis vaccines, diagnostic reagents and treatment drugs, and is of great significance in prevention and treatment of tuberculosis.

Description

technical field [0001] The invention belongs to the technical field of immunology, in particular to Mycobacterium tuberculosis-specific CD8 + T cell epitope peptide and its application. Background technique [0002] Tuberculosis is an infectious disease induced by Mycobacterium tuberculosis infection with the highest infection rate in the world, and its case fatality rate is second only to AIDS. In 2012 alone, there were 8.6 million new cases of tuberculosis worldwide and 1.3 million deaths. The new features of the tuberculosis epidemic are: the emergence and spread of drug-resistant tuberculosis cases; the co-infection of tuberculosis and AIDS is intensifying. For these two types of patients, there are currently no effective preventive measures. China is one of the countries with the heaviest burden of tuberculosis in the world, and it is time to develop tuberculosis vaccines and therapeutic drugs suitable for the Chinese population. [0003] Mycobacterium tuberculosis ...
