[0050] Experimental example 1 Intervention effect of rhein on Staphylococcus xylosus biofilm

[0051] 1. Experimental method

[0052] 1.1 Determination of the minimum inhibitory concentration (MIC, minimal inhibitory concentration) of rhein on Staphylococcus xylosus

[0053]The experimental operation and results were judged according to the standard microdilution method recommended by the National Committee for Clinical Laboratory Standards (NCCLS). Staphylococcus xylosus strains were inoculated on TSB agar plates and cultured overnight at 37°C. Take a single colony and inoculate it in TSB medium, and incubate at 37°C for 12h-16h. The concentration of the bacterial solution was adjusted to 0.5 McFarland tube suspension (1.0×10 8 cfu mL -1 ), and then diluted with THB medium to about 1.0×10 6 cfu mL -1 . Add TSB medium to the sterile 96-well cell culture plate in turn, except for the first well, add 160 μL TSB medium, add 100 μL TSB to each tube, add 40 μL rhein to the f...

Abstract

The invention discloses the application of rhein in preparing medicine for inhibiting a staphylococcus xylosus biofilm and belongs to the field of application of novel medicine of the rhein. According to the application, a staphylococcus xylosus biofilm in vitro model is adopted for researching the in vitro intervention effect of the rhein against the staphylococcus xylosus biofilm, through the crystal violet method and the observation with a scanning electro microscope, it is found that the rhein has a definite effect in inhibiting or intervening the formation of the staphylococcus xylosus biofilm, and the rhein can be applied to inhibiting staphylococcus xylosus, intervening the formation of the staphylococcus xylosus biofilm or exterminating the staphylococcus xylosus in the matured staphylococcus xylosus biofilm.

Description

technical field [0001] The invention relates to a new medical application of rhein, in particular to the application of rhein in the preparation of medicines for inhibiting Staphylococcus xylosus or Staphylococcus xylosus biofilm, and belongs to the field of new medical applications of rhein. Background technique [0002] Staphylococcus xylosus is a plasma coagulase-negative staphylococcus (CNS). This bacterium is widely distributed in nature and was previously considered to be a non-pathogenic bacterium. In recent years, plasma coagulase-negative staphylococci have been considered as emerging pathogens causing mastitis in dairy cows. Cow mastitis is one of the diseases that seriously harm the dairy industry, and it is also the third major disease that leads to the elimination of dairy cows. The most convincing argument that cow mastitis is difficult to cure and recurrent is its strong biofilm (Biofilm, BF) forming ability. Biofilm is a multi-complex matrix produced by ba...
