Problems solved by technology

When several konjac resources with the same flowering period are planted together or not far apart, pollination between different konjac resources is very frequent and easy under the pollination of flies or other insects, which seriously plagues konjac hybridization and directional selection new breed work

Embodiment Construction

[0018] Through experimental research, every morning is the best time for konjac artificial cross-pollination. When the konjac spathe opens naturally and its appendages emit a strong odor, it indicates that the pistil is mature, and cross-pollination can be carried out at this time. The specific operation is as follows:

[0019] The konjac stamen is cut off from the junction of the konjac stamen and the konjac pistil with a scalpel that is 75% alcohol sterilized by volume fraction before the dark of the day when the cross pollination of konjac is completed. Put the konjac stamens under the ground on the paper prepared in advance in a room with a room temperature of 20-25°C; the konjac stamens will release pollen in the next morning or a little longer, and when the konjac stamens release pollen, the pollen will naturally scatter on the paper above, collect the scattered konjac pollen from the paper, and use the collected konjac pollen immediately for cross-pollination of konjac ...

Abstract

The invention discloses an in-vitro collection method of konjac pollen. The method comprises steps as follows: a scalpel disinfected by 75% alcohol is used for cutting off konjac stamens from junctions of the konjac stamens and konjac pistils before darkness of the day when konjac hybrid pollination is finished; the cut konjac stamens are placed on an indoor paper prepared in advance; when the konjac stamens release pollen, the pollen naturally scatters on the paper, the scattered konjac pollen is collected from the paper and immediately applied to hybrid pollination of konjac plants with mature pistils or placed in a centrifugal tube, and the konjac pollen is stored in a refrigerator at the temperature of subzero 20 DEG C for pollination after being dried for 3 h under the vacuum condition. With the adoption of the method, problems that the amount of konjac pollen collected with a conventional method is small, operation is inconvenient and pollen mixing occurs among different kinds of konjac resources are solved, the collected konjac pollen per plant is 1.5-2 times that collected by the conventional method.

Description

technical field [0001] The invention belongs to the technical field of plant breeding, and in particular relates to a pollen collection method for plant breeding. Background technique [0002] Konjac (Amorphophallus rivieri) is a perennial herbaceous plant of the genus Amorphophallus Blume in the Araceae family. Its bulbs are rich in glucomannan (KGM). market prospects. With the continuous development of large-scale planting of konjac in my country and the continuous extension of the industrial chain, konjac has gradually become a characteristic industry for farmers in the mountainous and semi-mountainous areas of our country to get rid of poverty and become rich. [0003] Konjac has two reproductive methods: asexual tubers and sexual seeds. At present, people use asexual tubers to propagate seeds for a long time in production, resulting in a single genetic background, obvious degradation of species, decline in tuber quality and serious disease occurrence. The sustainable d...
