Problems solved by technology

The above-mentioned symbiotic bacteria are passed from mother to offspring in the vertical transmission mode of the host's ovary-oocyte in Hemiptera insects, and the position where these symbiotic bacteria enter the ovary is at the epithelial plug of the ovarian tubule , indicating that the epithelial plug of the ovarian tubule may have a unique location for the absorption of commensal bacteria, but whether this uniqueness is universal for the absorption of any foreign particles is unknown
In addition, there is a lack of controls to study the entry of symbionts into insect ovaries, which also hinders the study of the vertical transmission mechanism of insect symbionts through eggs

Embodiment

[0013] Example: Studying the method of absorbing red fluorescent particles by the ovary of the brown planthopper in vitro

[0014] The method proceeds as follows:

[0015] (1) Insect preparation: take mature female adults of N. lugens, disinfect the surface with 75% alcohol for 5 minutes, then wash with sterile 1×PBS (phosphate buffer saline, pH=6.4) for 3 times, and set aside;

[0016] (2) Obtaining isolated ovaries: Take 10 mature female adults of N. lugens in step (1), place them in sterile 1×PBS, and carefully dissect the abdomen of the worms with sterilized microdissecting forceps. In the case of destroying the intestines of the worms, the brown planthopper ovaries were obtained, and the brown planthopper ovaries were washed with 1 × PBS, and then 10 brown planthopper ovaries were placed in a small 35 mm diameter medium containing 1.5 mL IPL-41 (Giboco, 11405-081) insect medium. in a Petri dish;

[0017] (3) Co-cultivation of foreign particles and ovaries: red fluoresce...

Abstract

The invention discloses a method for carrying out in-vitro study on the absorption for foreign particles by the insect ovary. The method comprises the following steps: firstly dissecting an insect body to obtain the ovary, placing the ovary on a cell culture medium, adding the foreign particles on the cell culture medium, carrying out co-culture of the foreign particles and the insect ovary, and finally observing the condition that the foreign particles enter the insect ovary under a microscope. With the adoption of the method, the position specificity of the insect ovary for absorbing the foreign particles can be studied; and the foreign particles can be taken as the contrast of symbiotic bacteria, so that an experimental contrast is provided for the in-vitro study on the propagation of the symbiotic bacteria of the insects via a vertical propagation mechanism.

Description

technical field [0001] The invention relates to the technical field of insect physiology and biochemistry research, in particular to the field of physiology and biochemistry research of insect ovary. Background technique [0002] Insects are the most diverse animals in nature and are closely related to human production activities and health. Studying the physiological and biochemical characteristics of insects is the basis for utilizing or controlling insects. In some insects, especially Hemiptera insects that suck plant juice with piercing-sucking mouthparts, there are some symbiotic bacteria closely related to their survival and development. These symbiotic bacteria exist in the fat body of insects and provide the host with rice. Nutrients such as some essential amino acids that are scarce in the juice, without these symbiotic bacteria, insects will not be able to survive. For example, in brown planthoppers, there are a large number of yeast-like symbionts that provide e...
