[0009] Example Dyeing method of spider mites

[0010] At a temperature of 25°C-30°C and a humidity of 75%-85%, pour 30ml of 0.1% sodium fluorescein solution into a petri dish with a diameter of 5cm, stretch the parafilm, and cut out a circle with a diameter of 3cm , and then put the cut parafilm film flat on the surface of the sodium fluorescein solution in the petri dish so that the parafilm film floats on the petri dish solution, and finally place the spider mites that have been starved for 24 hours in a 2 ml centrifuge tube on the film In 2-4 hours, the spider mite digestive system can be observed under a fluorescent stereomicroscope.

Abstract

The invention relates to a dyeing method of a tetranychid digestion system. The dyeing method is characterized in that under the conditions of the temperature being 20 to 35 DEG C and the humidity being 75 to 90 percent, 0.05 to 5 percent sodium fluorescein or fuchsine solution is poured into a culture dish; then, the stretched parafilm is sheared into a round shape with the diameter being smaller than that of the culture dish; then, the sheared parafilm is flatly put onto the surface layer of the sodium fluorescein or fuchsine solution contained in the culture dish so that the parafilm floats on the solution in the culture disc; finally, tetranychid after the hungry treatment for 24 to 48 hours is put onto the parafilm; after 2 to 4 hours, the tetranychid digestion system can be observed under fluorescence or microscope. The dyeing method has the advantages that the operation is simple; the practical technical reference can be provided for observing the alimentary tract and the whole digestion system of the tetranychid.

Description

technical field [0001] The invention relates to a dyeing method for the digestive system of spider mites, belonging to the field of biological control of modern agricultural pests. Background technique [0002] At present, domestic research on spider mites is mainly limited to classification, breeding and application. There is little research on the microstructure of spider mites, especially the mechanism of the digestive system. [0003] The digestive system of spider mites is an important structure for the study of spider mites eating and digesting food. It is of great significance to observe the direction of food in the mites after spider mites eat food. . [0004] However, there is no report on how to observe the digestive system of spider mites conveniently. [0005] The purpose of the present invention is to provide a simple and efficient staining method for the digestive system of spider mites without causing the death of mites.
