Embodiment 1

[0024] The indirect ELISA kit for detecting serum type 4 avian adenovirus antibody based on the F2 protein of the present invention can be obtained by the following steps:

[0025] (1) Isolation of FAdV-4 avian adenovirus:

[0026] Take the liver of a dead chicken suspected of avian adenovirus infection, grind it and add PBST at a ratio of 1:5 to make a suspension; centrifuge at 5000r / min for 15min, and take the supernatant; React at 37°C for 30 minutes; filter through a 0.45um microporous filter, inoculate 8-day-old SPF chicken embryos through the allantoic cavity at a dose of 0.2ml / embryo, collect allantoic fluid 96-120 hours after inoculation; After being identified as FAdV-4 avian adenovirus, it was stored at -20°C.

[0027] (2) Preparation of avian adenovirus genome:

[0028] Take 400uL supernatant of FAdV-4 isolated strain virus (serotype 4 avian adenovirus) in a 1.5mL finger tube, add 400uL cell lysate (50mmol / L Tris-HCl pH 8.0, 20mmol / L EDTA pH 8.0, 2% SDS and prote...

Abstract

The invention discloses an F2 protein based indirect ELISA kit for detection of fowl adenovirus serotype 4 (FAdV-4) antibody. The kit includes an ELISA plate coated with a spike protein gene expression product, a positive and negative control, an HRP labeled rabbit anti-chicken secondary antibody, a sample diluent, a color development liquid and a washing liquid. The indirect ELISA kit for FAdV-4antibody established by the invention can specifically detect the FAdV-4 antibody, and does not react with anti-influenza virus (AIV) serum, anti-mardivirus (MDV) serum, anti-Newcastle disease virus (NDV) serum, anti-avian reticuloendotheliosis virus (REV) serum and SPF chicken serum (Figure 6). Therefore, the kit has good FAdV-4 specificity, and can be used for FAdV-4 infection and immune statusepidemiological investigation.

Description

technical field [0001] The invention belongs to the field of biotechnology detection, and in particular relates to an indirect ELISA kit for detecting serum type 4 avian adenovirus antibody based on F2 protein. Background technique [0002] Fowl Adenovirus (Fowl Adenovirus, FAdV) belongs to Adenoviridae, and is divided into 5 species (A-E) and 12 serotypes. Although it has been reported all over the world, FAdV infection generally causes subclinical conditions, while acute infection mainly causes inclusion body hepatitis, pericardial effusion, and gizzard erosion. Since 2013, cases of inclusion body hepatitis and pericardial effusion caused by FAdV in domestic chicken flocks have gradually increased. By 2015, FAdV infection broke out in chicken flocks in several provinces in China. At present, the outbreak of FAdV in China not only occurs in 3-4 week-old broilers, but also occurs in 10-20 week-old laying hens, causing serious economic losses to the chicken industry. Virus...
