[0039] 1. Test material

[0040] (1) Plant material: seeds of two species (YK, YD) of Cymbidium mannii and Cattleya hybrida.

[0041] (2) Fungal materials: 7 strains of mycorrhizal fungi (Tulasnellas pp.) of the genus Orchidaceae obtained from experiments in our laboratory for many years, see Table 1.

[0043] (1) Configuration of culture medium

[0044] The configuration of symbiotic medium (oat medium OMA): take oatmeal and grind it into powder, take three gradients of oat flour respectively, and prepare 0.5g / L, 1.0g / L, 2.0g / L, 3.0g / L , 4.0g / L, 8.0g / L, 12.0g / L OMA solutions. Boil in deionized water for 1 hour, filter, add 8.0 g of agar, add deionized water to the filtrate to 1000 mL, and sterilize at high temperature.

Abstract

The invention discloses a method for screening tulasnella fungi and establishing a symbiotic system for promoting seed germination of cymbidium mannii and cattleya hybrida. A Tcs1 bacterial strain isadopted and establishes a symbiotic relation together with one or more orchid seeds including cymbidium mannii seeds, cattleya hybrid YK seeds and cattleya hybrid YD seeds to promote germination of orchid seeds. A symbiotic germination test is carried out on an artificial medium through adoption of tulasnella fungi and orchid seeds, and is compared with a control group of orchid seeds with no fungi inoculated to screen mycorrhizal fungi capable of widely efficiently promoting seed germination and to screen appropriate symbiotic mediums. Efficient symbiotic fungi for a germination stage of orchid seeds are obtained, the germination rate of orchid seeds is improved via symbiotic germination culture technique, and a symbiotic system of orchid and mycorrhizal fungi is established.

Description

technical field [0001] The invention relates to a biological technology, in particular to a method for establishing a symbiotic system of a fungus belonging to the genus Colloides which effectively promotes the germination of sclerophyllum and cattleya seeds. Background technique [0002] Orchidaceae (Orchidaceae) belongs to the class Monocots of the angiosperm phylum, and there are about 700 genera and more than 35,000 species distributed in the world (Dressler, 1993; Cribb, 2003), and its number of species ranks second only to Compositae. Branch (Lang Kaiyong, 1994). It is beautiful in shape, bright in color, and has great ornamental and medicinal value. [0003] Orchids have two notable characteristics: [0004] 1. The embryo of Orchidaceae seeds is only composed of dozens of cells, which is incompletely differentiated and difficult to germinate naturally. [0005] 2. Orchidaceae are almost all mycorrhizal plants. In the natural growth state, no matter green leaf orchi...
