Abstract

The invention provides a preparation method and application of a cell combination preparation for treating diabetes mellitus type I, and belongs to the field of regenerative medicine and diabetology.The preparation method comprises the following steps: step A. obtaining tissues of human amniotic epithelial cells (hAECs); step B. carrying out constant temperature culture in vitro; step C. inducingdifferentiation and collecting cells; step D. subculturing; step E. preparing a hyaluronic acid solution; step F. adding the hAECs cells to the hyaluronic acid solution and adjusting; and step G. carrying out intravenous injection and transplanting. Animal experiments can clearly show that the drug combination model group can greatly improve the blood glucose level and insulin level in a mouse body, and after the drug combination model group is transplanted for eight weeks, the numerical values of the blood glucose level, insulin level and abdominal cavity glucose tolerance in the mouse bodyare very close to the numerical values of the normal group; the level of inflammatory factors in plasma is significantly down-regulated; and in summary, based on the good biological characteristics ofhAECs and hyaluronic acid, a method of treating the diabetes mellitus type I through combination of the hAECs and the hyaluronic acid transplantation is established for the first time, and significant curative effects are achieved.
