Abstract

The invention belongs to the technical field of low-temperature storage of bananas and discloses an in-vitro banana stem tip ultralow-temperature storage method comprising the steps: pre-culturing a banana seedling with the height of 3.0-6.0 cm in a culture medium containing 0.4 mol / L of sucrose for 2 d; stripping off a stem tip with 1-2 leaf primordia under the condition of room temperature; loading the stem tip in a loading liquid, then, treating the stem tip by using a vitrification solution, and preparing droplets on an aluminum foil strip; storing the stem tip in liquid nitrogen for at least 1 h, defrosting and washing the stem tip by using 1.2 mol / L of sucrose culture solution, wherein the stem tip is washed for 10 min every time; and transferring the treated stem tip to an MS culture medium containing 0.3 mol / L of sucrose, culturing the stem tip in a dark room for 2 d, then, transferring the stem tip into an MS culture medium containing 0.5 mg / L of 6-BA, culturing the stem tip for 30-35 d, inoculating seed sources recovered to normally grow into a novel recovery growth culture medium for multiplication and rapid propagation to obtain cluster buds, and carrying out culture ina rooting culture medium.
