Problems solved by technology

In our previous study, a small dose (10 μg) of chicken egg albumen (OVA) was challenged by intranasal drops, and successfully replicated a mouse EB model without airway hyperresponsiveness (AHR) and airway eosinophilic inflammation (invasive airway reactivity measurement), but the model has the following shortcomings: ① Cough is an important clinical feature of EB, and some asthmatic patients also show cough, but this study failed to study cough in mice; ② The therapeutic effect of hormones on cough was not observed; ③This method (i.e., invasive airway responsiveness measurement) will kill the mice immediately after the test is completed, which does not meet the ethical requirements and cannot produce qualified models in batches, which is time-consuming and labor-intensive. In experiments that require animals to survive, the current application of this model is limited and it is difficult to promote

[0042] 1. Experimental animals and grouping

[0043]SPF-grade BALB / c mice, female, 7 weeks old, weighing 18-20g, were purchased from the Experimental Animal Center of Southern Medical University; the mice were all kept in the SPF-grade animal room of the State Key Laboratory of Respiratory Diseases, and the day and night alternated between light and dark for 12 hours / 12h, free to eat feed and triple distilled water without OVA (egg protein, Sigma, USA). 24 mice were randomly divided into 4 groups (6 in each group): saline group (NS group), asthma group (AS group), eosinophilic bronchitis group (EB group), dexamethasone treatment group (DXM Group).

[0044] (1) Sensitization: AS group, EB group, and DXM group were sensitized by intraperitoneal injection of 0.2ml OVA solution (containing OVA 10μg + adjuvant aluminum hydroxide 0.1ml) on the 0th day, the 7th day, and the 14th day. The same dose of normal saline was injected into the intraperitoneal cavity of the group. During t...

Abstract

The invention discloses a method for establishing and detecting a mouse model of eosinophilic bronchitis. The method comprises the following steps: (1) sensitizing and stimulating the mice with OVA to establish a mouse model of eosinophilic bronchitis; (2) placing the mice obtained in step (1) in a Buxco non-invasive lung function Free movement in the closed body scanning chamber of the detection system; among them, the Buxco non-invasive pulmonary function detection system is connected with an atomization controller and a signal converter; (3) mice are stimulated by nebulization with methacholine, and the gas flow through Finepointe software is used to stimulate the mice. The airway responsiveness of the mice was evaluated by measuring the change of Penh value before and after nebulization challenge in the airway responsive mode. The invention establishes a mouse EB model with obvious cough, no airway hyperresponse, airway eosinophilic granulocyte inflammation, effective hormone therapy and conforming to ethics, which is beneficial to popularization and application of the mouse EB model.

Description

technical field [0001] The invention belongs to the field of medical research, in particular to a method for establishing and detecting a mouse model of eosinophilic bronchitis. Background technique [0002] At present, most of the researches on eosinophilic bronchitis (EB) focus on the clinic, but in view of the limitations of human studies, the exploration of EB needs to be carried out to a large extent through animal experiments, and there is currently no animal model of EB, so it is successful. It is of great significance to establish an EB animal model. At present, no exact EB animal model has been established in the world. Only the Japanese scholar Ogawa proposed the method of establishing a model of EB guinea pigs by intranasal injection of polymyxin B. Pathological conditions, high feeding costs, few biological agents, etc.), no other scholars have used this model to study EB. [0003] Mice share 99% of their genes with humans, and the relevant detection reagents a...
