Problems solved by technology

However, there are no large-scale studies using immune-related genes to predict the prognosis of colorectal cancer
[0005] The main disadvantage of the existing technology: there is no organic combination of the role of immune-related genes in colorectal cancer, and no large-scale validation
More importantly, there are many problems in the use of existing gene combinations. For example, many products require the use of a complete set of kits, and patients can only be predicted by re-measurement under the premise of complete standards, and there is no compatibility with other measurement methods.

Embodiment 1

[0019] Discovery of prognostic immune-related genes in colorectal cancer

[0020] The data set numbered GSE39582 in the public high-throughput GEO database is used as the development data set, and the measurement platform is the microarray platform of Affymetrix Company, which contains 566 samples of colorectal cancer patients, hereinafter referred to as the CIT microarray data set. Among them, there were 557 patients with complete clinical prognosis information, and 309 cases of early colorectal cancer. The immune-related genes were obtained from the ImmPort database, with a total of 1811 genes and 17 categories, of which 1264 immune-related genes were available in the GSE39582 dataset.

[0021] Construction of immune-related genes for prognosis prediction of colorectal cancer

[0022] According to the patient's prognosis information, using the LASSO Cox model, the 1264 immune-related genes were reduced to 23 genes, such as figure 1 In addition, see Table 1 for the list...

Embodiment 2

[0028] Example 2 Prediction of colorectal cancer prognosis using this model

[0029]Tested on a training cohort (CIT / GSE39582), two validation cohorts (TCGA CRC and Meta-validation), a total of 1587 patient samples. As shown in the figure, the test effect on the CIT microarray data set (HR=2.55, 95%CI=1.88-3.45, P<0.001), on the colorectal cancer patients in the TCGA CRC (validation-1) data set The test effect (HR=1.64, 95%CI=1.10-2.44, P=0.001), the test effect (HR =2.61, 95%CI=1.71-3.98, P<0.001).

[0030] As shown in Table 2, the present invention also compares the constructed immune signature gene model (IRGS) with the Oncotype DX algorithm model proposed internationally at present; the results found that in the CIT / GSE39582 and TCGA CRC cohorts, the prediction index of IRGS (C- index) are better than the Oncotype DX algorithm, and the prediction index in the Meta-validation verification data set is equal to that of the Oncotype DX algorithm.

Abstract

The invention provides an application of an immune-related gene in a kit and a system for predicting the prognosis of a colorectal cancer. A group of characteristic genes capable of stably predictingthe prognosis of the colorectal cancer is found by combining the immune-related gene; on the premise that a standardized technology platform is not adopted to uniformly measure, any type of measurement platform can be compatible to predict the prognosis of the colorectal cancer; by adopting the system, the prognosis of colorectal cancer can be well predicted and the risk of early colorectal cancerpatients can be remarkably predicted. Univariate and multivariate analysis can also be carried out, and it is proved that the prognosis risk of colorectal cancer patients can be independently predicted by using an immune risk model (IRGS).

Description

technical field [0001] The invention relates to the application of immune-related genes in a kit and system for the prognosis of colorectal cancer. Background technique [0002] Colorectal cancer (CRC) is one of the most common cancers in the world, with nearly 1.4 million new cases each year. Although new treatment options emerge in an endless stream, the 5-year survival rate is only 55%. Surgical treatment is the first-line option, but according to the traditional clinical characteristics, patients of the same type have very different prognosis after treatment. In recent years, studies have suggested that this is mainly caused by the molecular heterogeneity of cancer patients. [0003] Gene molecular markers refer to the establishment of mathematical models based on the expression of a group of genes through machine learning to predict specific clinical targets. In recent years, gene expression detection methods have been quite mature, including high-throughput RNA sequ...
