Abstract
The invention discloses a regeneration method of 'Da-ding-xiang' litchi variety in vitro. The regeneration method of the 'Da-ding-xiang' litchi variety in vitro comprises the following steps: 1) carrying out callus cultivation by taking litchi anther as an explant so as to obtain embryogenic calluses; 2) performing somatic embryo induction culture on the embryogenic calluses by using a medium, which consists of IAA, ABA, 6-BA, inositol, lactoalbumin hydrolysate and coconut water, so as to obtain somatic embryos; 3) performing maturation culture on the somatic embryos by using a medium, which contains coconut water, so as to obtain mature somatic embryos; and 4) performing regeneration culture on the mature somatic embryos by using a medium, which contains 6-BA and GA3, so as to obtain regenerated plants. According to the invention, a culture method of the regenerated plants of the 'Da-ding-xiang' litchi variety in vitro is established so as to achieve very high somatic embryo inductionrate and germination rate, so that the culture method is capable of improving production efficiency and reducing cost; and thus, a good foundation is laid for litchi variety improving and breeding bybiotechnology.
