Embodiment 1

[0032] The construction method of embodiment 1 preeclampsia mouse animal model

[0033] 1. Construction strategy of Mettl3 conditional gene knockout mice

[0034] The Mettl3 conditional gene knockout mouse construction strategy is as follows: figure 1 : The mouse Mettl3 gene is located on chromosome 14 and encodes 11 exons. Loxp1 and Loxp2 are inserted outside of exons 2 and 3, respectively, and exons 2 and 3 of the Mettl3 gene to be knocked in the middle. In the presence of recombinant Cre enzyme, the sequence between the two Loxp sites was knocked out.

[0035] (1) According to the location of the Mettl3 gene to be knocked out, order BAC clones RP23-137N7 (Saiye Biotech) and RP23-277J4 (Saiye Biotech) to construct targeting vectors;

[0036] (2) Transforming the linearized targeting vector into mouse ES cells, and screening positive ES cells;

[0037] (3) Microinject the positive ES cells into mouse blastocysts, and transplant the ES cell-injected blastocysts into the ute...

Abstract

The invention discloses a construction method and application of a preeclampsia mouse animal model. According to the construction method, the m6A modification level in a mouse is reduced, and a mutant mouse with the reduced RNA m6A modification level, namely the preeclampsia mouse animal model, is obtained; specifically, a Mett13<+/>hybrid mutant mouse is constructed through an ES targeting technology, and a result shows that the sensitivity of the mutant mouse to a PE inducer L-NAME is increased, the mutant mouse presents the characteristics of PE, and a new thought and a new means are provided for etiological diagnosis, prevention and treatment of preeclampsia.

Description

technical field [0001] The present invention relates to the technical field of disease animal model construction, and more specifically, relates to a method for constructing a preeclampsia mouse animal model and its application. Background technique [0002] Preeclampsia (PE) pregnant women develop new hypertension after 20 weeks of pregnancy, accompanied by proteinuria or edema, dizziness, etc., and in severe cases, they may be accompanied by systemic multi-organ lesions. The global incidence of preeclampsia is 5% to 10%, and its incidence can be as high as 15% to 20% in areas with inadequate perinatal care. It is an important cause of maternal death, premature birth and neonatal death. one. The pathogenesis of preeclampsia is still unclear, but current studies suggest that its occurrence is closely related to early trophoblast invasion disorder and spiral artery remodeling disorder. [0003] Due to the limitations of medical ethics, animal models have become an important...
