Embodiment 1

[0036] This experiment has been approved by the Ethics Committee of Xuanwu Hospital, Capital Medical University.

[0037] Animals were housed in SPF chambers and had free access to food.

[0038] The primary culture of rat bone marrow stromal cells was obtained from male donor Sprague-Dawley rats (4 weeks old, 150-200g), and BMSCs were isolated. For specific methods, please refer to the following literature: Zhao Chunsong, Zou Haiqiang, Yan Xiaoming, Guan Yunqian, Chen Ling, Wang Jiayin, Zhang Yu. Inhibition of human umbilical cord mesenchymal stem cells on the proliferation of peripheral blood lymphocytes in patients with Parkinson's disease. Journal of Capital Medical University. June 2014 No. 35(3):344-352.).

[0039] At the time point of day 2, 30 male Sprague-Dawley young mice (8 weeks old, 280-320 g) were randomly divided into sham operation group, ischemia group, ischemia+rMSCs group (n=10 in each group). In each group, animals were used for immunofluorescence staining...

Abstract

The invention provides a kit and a method for evaluating the curative effect of mesenchymal stem cells on cerebral infarction, and belongs to the technical field of medicines. The kit comprises a reagent for detecting the number of astrocytes, microglia, mononuclear cells, neutrophils and lymphocytes in a cerebral cortex infarction region and a reagent for detecting the expression quantity of TNF-alpha in the cells. The method comprises the following steps: respectively detecting the numbers of astrocytes, monocytes and lymphocytes and the expression quantity of TNF-alpha in a cerebral cortex infarction region before and after the mesenchymal stem cells are transplanted, and if the numbers of the astrocytes, the monocytes and the lymphocytes are lower than those before the mesenchymal stem cells are transplanted, and the expression quantity of TNF-alpha in astrocytes, mononuclear cells and lymphocytes is lower than that before transplantation, determining that the curative effect is good. The kit and the method can efficiently and accurately evaluate the curative effect of the mesenchymal stem cells on cerebral infarction.

Description

technical field [0001] The invention belongs to the technical field of medicine, and in particular relates to a kit and a method for evaluating the curative effect of mesenchymal stem cells in treating cerebral infarction. Background technique [0002] Cerebral infarction, formerly known as cerebral infarction, also known as cerebral ischemic stroke, refers to the localized ischemic necrosis or softening of brain tissue caused by cerebral blood supply disorder, ischemia and hypoxia. Cerebral infarction includes common cerebral arteriosclerosis thrombotic cerebral infarction (referred to as cerebral thrombosis) and cerebral embolism. Cerebral infarction is a common ischemic cerebrovascular disease in the elderly, and it can make most patients disabled, and the loss of labor force causes many inconveniences in life. [0003] Cerebral infarction needs to be treated in time, and the choice of treatment method is extremely critical. The traditional treatment methods for cerebra...
